目的:探讨结肠癌细胞株SW-480中脂联素受体的表达情况以及人脂联素重组体对结肠癌体外增殖侵袭能力的影响。方法通过RT-PCR试验检测脂联素受体1和受体2在结肠癌细胞株中的表达,然后将SW-480细胞分设对照组和脂联素重组体作用(1、10、30、45、60μg/mL)组,作用后,分别以MTT法、划线迁移实验检测脂联素重组体对结肠癌细胞增殖、迁移能力的影响。结果脂联素受体1、2在该细胞株中均有表达,脂联素重组体质量浓度高于10μg/mL时,对SW-480细胞生长具有明显的抑制作用(P<0.01),抑制作用随着浓度增高而增强;当脂联素重组体浓度大于30μg/mL时,能明显地降低肿瘤细胞体外侵袭迁移能力(P<0.01),抑制率随脂联素重组体浓度的升高而增加,介于1.8%~42.0%之间。结论脂联素重组体在大于10 g/mL时,可能对结肠癌细胞的增殖迁移产生较为明显的影响。

목적:탐토결장암세포주SW-480중지련소수체적표체정황이급인지련소중조체대결장암체외증식침습능력적영향。방법통과RT-PCR시험검측지련소수체1화수체2재결장암세포주중적표체,연후장SW-480세포분설대조조화지련소중조체작용(1、10、30、45、60μg/mL)조,작용후,분별이MTT법、화선천이실험검측지련소중조체대결장암세포증식、천이능력적영향。결과지련소수체1、2재해세포주중균유표체,지련소중조체질량농도고우10μg/mL시,대SW-480세포생장구유명현적억제작용(P<0.01),억제작용수착농도증고이증강;당지련소중조체농도대우30μg/mL시,능명현지강저종류세포체외침습천이능력(P<0.01),억제솔수지련소중조체농도적승고이증가,개우1.8%~42.0%지간。결론지련소중조체재대우10 g/mL시,가능대결장암세포적증식천이산생교위명현적영향。
Objective To investigate the expression of adiponection receptor(adipoR) and the effects of recombinant adiponectin (gAd) on the proliferation and migration of human colon cancer cell line SW-480.Methods SW-480 cells were divided into control group and gAd(1, 10, 30, 45and 60μg/mL) treatment groups, the effect of gAd on proliferation of SW-480 cells was measured by MMT assay, the invasive and migration abilities were assayed by scratch test.Results The proliferating ability of the cell was inhibitied in a concentration-dependent manner, with the ranging from 2.9%-37.6%, the cell proliferation was dramatically inhibited when the concentration of gAd was higher than 10μg/mL (P<0.01); the migration were also inhibitied in a concentration-dependent manner, with the ranging from 1.8% - 42.0%, the migration ability of SW-480 cells were dramatically inhibited when the concentration of gAd was higher than 30μg/mL (P<0.01).Conclusion These results indicate that the adiponectin dramatically inhibited the proliferation and migration of colon tumor cell when its concentration is >10μg/mL.
