[目的]观察肺瘤平膏对肺癌A549细胞增殖作用的影响和炎性肿瘤微环境下A549细胞迁移和侵袭能力的影响。[方法]40只Wistar大鼠随机分成5组,分别给予生理盐水、肺瘤平膏不同剂量灌胃及环磷酰胺腹腔注射,2次/d,给药3d后,取血,制备含药血清;通过MTT法观察肺瘤平膏不同剂量对A549细胞增殖的影响;建立A549细胞与巨噬细胞的共培养体系,观察炎性肿瘤微环境对A549细胞形态的影响及肺瘤平膏的干预作用;通过Transwell实验观察肺瘤平膏对炎性肿瘤微环境下A549细胞迁移和侵袭能力的影响。[结果]各含药血清组均在药物干预24h后出现抑制作用,48h后抑制作用最强,72h后抑制率又有所下降。共培养条件下,A549细胞变成间充质细胞样,而加入肺瘤平膏含药血清组,部分A549细胞形态恢复上皮细胞样。Transwell迁移和侵袭实验结果表明,各组含药血清均能降低炎性肿瘤微环境下A549细胞迁移和侵袭能力,其中肺瘤平膏中剂量组效果最好。[结论]肺瘤平膏可以抑制A549细胞的增殖,并能降低炎性肿瘤微环境下A549细胞的迁移和侵袭能力。

[목적]관찰폐류평고대폐암A549세포증식작용적영향화염성종류미배경하A549세포천이화침습능력적영향。[방법]40지Wistar대서수궤분성5조,분별급여생리염수、폐류평고불동제량관위급배린선알복강주사,2차/d,급약3d후,취혈,제비함약혈청;통과MTT법관찰폐류평고불동제량대A549세포증식적영향;건립A549세포여거서세포적공배양체계,관찰염성종류미배경대A549세포형태적영향급폐류평고적간예작용;통과Transwell실험관찰폐류평고대염성종류미배경하A549세포천이화침습능력적영향。[결과]각함약혈청조균재약물간예24h후출현억제작용,48h후억제작용최강,72h후억제솔우유소하강。공배양조건하,A549세포변성간충질세포양,이가입폐류평고함약혈청조,부분A549세포형태회복상피세포양。Transwell천이화침습실험결과표명,각조함약혈청균능강저염성종류미배경하A549세포천이화침습능력,기중폐류평고중제량조효과최호。[결론]폐류평고가이억제A549세포적증식,병능강저염성종류미배경하A549세포적천이화침습능력。
Objective] To observe the effect of Fei-Liu-ping ointment on lung cancer A549 cells proliferation and migration, invasion under inflammatory tumor microenvironment. [Methods] Forty Wistar rats were randomly divided into five groups, given saline, different doses of Fei-Liu-Ping ointment and CTX, twice a day. All the procedures continued for 3 days. blood was collected to prepare Fei-Liu-Ping ointment-containing serum(FLP); To observe the effect of FLP on lung cancer cell proliferation by MTT; To establish co-culture system of A549 cells and macrophage and to observe the effect of inflammatory microenvironment on A549 cells and to observe what roles FLP played in inflammatory microenvironment; To observe the effect of FLP on A549 cell migration, invasion under inflammatory tumor microenvironment by Transwell. [Results] FLP treatment inhibited A549 cell proliferation after 24h, which was stronger after 48h, and the inhibition rate decreased after 72h; A549 cells developed into mesenchymal-like cells in co-culture conditions, which could be blocked by FLP in part; For Transwell migration and invasion experiment, FLP treatment reduced the cell numbers of A549 cells under inflammatory tumor microenvironment. [Conclusions] FLP could inhibit A549 cell proliferation and inhibit A549 cell invasion and migration under inflammatory tumor microenvironment.
