目的 探索苯职业接触者细胞色素氧化酶P450 2E1 (CYP2E1)Rsa Ⅰ/Pst Ⅰ与Dra Ⅰ基因多态性.方法 采用横断面调查的方法,选择71名苯接触人群为观察组,以71名无职业性苯接触的人员为对照组.采集两组研究对象血样,应用限制性片段长度多态性聚合酶链反应(PCR-RFLP)方法对其CYP2E1 Rsa Ⅰ/Pst Ⅰ与Dra Ⅰ基因多态性进行研究分型.结果 观察组与对照组CYP2E1 Dra Ⅰ位点基因型和等位基因的分布差异无统计学意义(基因型x2=2.374,P>0.05;等位基因x2=2.113,P>0.05);观察组与对照组CYP2E1 Rsa Ⅰ/Pst Ⅰ位点的基因型与等位基因分布差异均有统计学意义(基因型x2=9.129,P<0.01;等位基因x2=6.028,P<0.05).结论 CYP2E1 Rsa Ⅰ/Pst Ⅰ位点突变可增强体内CYP2E1的表达,提示携带该基因的个体对慢性苯中毒的易感性升高.

Objective To investigate the cytochrome P450 2E1 (CYP2E1) RsaⅠ/PstⅠ and DraⅠ polymorphisms in workers exposed to benzene.Methods A cross-sectional survey was carried out.A total of 71 workers exposed to benzene were included in observation group and the same number of people without occupational benzene exposure were included in control group.Blood samples from the two groups were collected and genotyping for CYP2E1 RsaⅠ/PstⅠ and DraⅠ were conducted using the polymerase chain reactionrestriction fragment length polymorphism.Results There were no significant differences in CYP2E1 DraⅠ genotype and allele distributions between the observation group and the control group (x2=2.374,P>0.05; x2=2.113,P>0.05).Significant differences in CYP2E1 RsaⅠ/PstⅠ genotype and allele distributions between the two groups were observed (x2=9.129,P<0.01; x2=6.028,P<0.05).Conclusion Mutations at CYP2E 1 RsaⅠ/PstⅠ can enhance the expression of CYP2E1 and this suggests individuals with the mutated gene have increased susceptibility to chronic benzene poisoning.
