目的:探讨PPARγ配体在肿瘤放射治疗中对肿瘤细胞的作用。方法采用RT-PCR方法检测CT-26细胞中是否有PPARγ表达。采用MTT方法检测PPARγ配体(吡格列酮)对CT-26细胞生长的影响。采用Western blot方法观察10 Gy X射线对CT-26细胞相关因子COX-2,PAI-1表达的影响,分析PPARγ配体(吡格列酮)对放射后CT-26细胞中COX-2、PAI-1表达变化的影响。结果 PCR结果显示CT-26细胞中有PPARγ表达。 MTT结果显示:吡格列酮可以降低CT-26细胞的存活率,10 Gy照射+吡格列酮处理48 h时CT-26细胞存活率降低更为明显。10 Gy X射线照射CT-26细胞后Western blot方法检测结果显示:照射后COX-2表达水平升高,照射后24 h COX-2表达水平最高,吡格列酮对照射诱导的COX-2水平升高无明显影响。10 Gy X射线照射CT-26后Western blot方法检测结果显示:照射后24 hPAI-1表达水平明显增高,吡格列酮对照射诱导的PAI-1表达水平升高起降低作用。结论吡格列酮等噻唑烷二酮类药,作用于小鼠结肠癌细胞,在肿瘤放疗过程中可能发挥潜在的协同作用。

Objective To observe the effect of PPARγligand ( Pioglitazone) radiotherapy on mouse colonal cancer cells (CT-26).Methods After CT-26 cell was radiated by 10 Gy X-ray,RT-PCR was used to detect PPARγgene expression.MTT assay was used to assay the growth of CT-26 cell after the treatment by PPARγligand ( Pioglitazone) ,irradiation and the 2 com-bined.Western blot methods were used to measure the expression of COX-2,PAI-1 after CT-26 cell was irradiated by 10 Gy X-ray and treated by pioglitazone in different time.Results ①RT-PCR results indicated that PPARγmRNA was expressed in mouse colon cancer cell line.②MTT results showed that pioglitazone inhibits CT-26 growth,the influence was obvious after treated by pi-oglitazone +10 Gy X-ray radiation.③Western blot results indicated that expression of COX-2 and PAI-1 protein in mouse colon cancer cell line increased after irradiated by 10 Gy X-ray.Pioglitazone had no influence on the level of radiation-induce COX-2 overexpression,but decreased the level of radiation-induced PAI-1 overexpression after CT-26 cell was treated by 10 Gy X-ray ir-radiation for 24 h.After CT-26 cell was irradiated by X-ray,the expression of PAI-1,COX-2 increased.PPARγligand ( pioglata-zone) decreased the expression of radiation-induced PAI-1 but had no influence in the level of radiation-induce COX-2.Conclu-sion Thiazolidinedione,such as pioglatazone,plays potential benefit role to cancer patients during radiation.
