目的：推理选育他克莫司高产菌株。方法在研究葡萄糖对他克莫司产生菌影响的基础上，应用葡萄糖的结构类似物2－脱氧－D－葡萄糖对他克莫司产生菌进行抗性选育。结果获得发酵水平较原始出发菌株FCZ0311－7提高19％的突变株7－12，该菌株遗传稳定。结论利用2－脱氧－D－葡萄糖对他克莫司产生菌进行推理选育可行、高效。
목적：추리선육타극막사고산균주。방법재연구포도당대타극막사산생균영향적기출상，응용포도당적결구유사물2－탈양－D－포도당대타극막사산생균진행항성선육。결과획득발효수평교원시출발균주FCZ0311－7제고19％적돌변주7－12，해균주유전은정。결론이용2－탈양－D－포도당대타극막사산생균진행추리선육가행、고효。
OBJECTIVE To obtain tacrolimus high producing strain using rational screen strategy.METHODS Use 2-deoxy-D-glucose,the structural analogue of glucose,to screen high production mutant on the base of studying effect of glucose on the tacrolimus producing strain.RESULTS A high production mutant,strain 7-12,was obtained and the relative fermentation titer of tacrolimus was about 19% higher than that of the original strain,Streptomyces sp.FCZ0311-7.CONCLUSION Rational screen tacrolimus producing strain by 2-deoxy-D-glucose resistance was feasible and efficient.
