Ethanol Extract of Zingiber%Prostatic cancer%Proliferation%PC-3 cells
目的:观察生姜醇提取物(extract of zingiber ethanol,EZE)对人前列腺癌 PC-3细胞体外增殖的影响,并探讨其可能的调控机制。方法运用 MTT 法检测不同浓度 EZE 对 PC-3细胞体外增殖的影响;Western-blot 检测不同浓度 EZE 作用于 PC-3细胞一定时间后其 P53、Bcl-2蛋白的表达情况。结果MTT 法结果提示,随着 EZE 浓度的增加与作用时间的延长,PC-3细胞 DE 增殖抑制率均增加,与对照组比较均有显著差异(P<0.05);Western-blot 结果显示 EZE 作用 PC-3细胞后可上调 P53、下调 Bcl-2蛋白的表达,并呈一定的量效关系。结论EZE 能够抑制 PC-3细胞体外增殖,其机制可能与其上调 P53、下调 Bcl-2蛋白的表达有关。

목적:관찰생강순제취물(extract of zingiber ethanol,EZE)대인전렬선암 PC-3세포체외증식적영향,병탐토기가능적조공궤제。방법운용 MTT 법검측불동농도 EZE 대 PC-3세포체외증식적영향;Western-blot 검측불동농도 EZE 작용우 PC-3세포일정시간후기 P53、Bcl-2단백적표체정황。결과MTT 법결과제시,수착 EZE 농도적증가여작용시간적연장,PC-3세포 DE 증식억제솔균증가,여대조조비교균유현저차이(P<0.05);Western-blot 결과현시 EZE 작용 PC-3세포후가상조 P53、하조 Bcl-2단백적표체,병정일정적량효관계。결론EZE 능구억제 PC-3세포체외증식,기궤제가능여기상조 P53、하조 Bcl-2단백적표체유관。
Objective To investigate the effects of ethanol extracts from Zingiber (EEZ) on the proliferation of human pro-static cancer PC-3 cells (PC-3) and its possible mechanisms. Methods MTT was used to analyze the effects of different doses of ethanol extract of Zingiber on the proliferation of human Prostatic cancer PC-3 cells. Western-blot was applied to detect the expressions of bcl-2 and p53 protein in PC-3 cells after treated by EEZ. Results MTT assay showed that the inhibition rate of cell proliferation all increased along with the increase of concentration and interaction time of EEZ. But it was sig-nificantly higher in the experimental group than in the control group (P<0.05). Western-blot assay showed that P53 protein expression increased while Bcl-2 protein expression reduced within a certain range of treating time and dose in PC-3 cells treated by EEZ. Conclusion Ethanol extract of Zingiber could inhibit the proliferation of PC-3 cells in vitro within a certain range of treating time and dose. The increase of P53 protein expression and decrease of Bcl-2 protein expression might be involved.
