目的::探讨牙龈间充质干细胞(GMSCs)复合脱细胞真皮基质(ADM)修复皮肤缺损的实验。方法:以纯化3代 GMSCs 作为种子细胞接种于 ADM,构建组织工程化皮肤替代物。实验分组:18只 SD大鼠随机分成3组:GMSCs 复合 ADM 移植组、单纯 ADM 组和空白对照组。将3组相应材料分别移植于 SD 大鼠背部皮肤缺损处,观察比较创面修复效果。结果:GMSCs 复合 ADM 移植组、单纯 ADM 组和空白对照组皮肤缺损创面愈合时间分别为(15.1±1.3)d、(18.5±2.1)d 和(22.6±2.6)d,差异有显著性意义(P <0.05)。组织学观察显示,GMSCs 复合 ADM 移植组成纤维细胞、新生小血管、胶原纤维均较单纯 ADM 移植组和空白对照组明显增多。结论:GMSCs 复合 ADM 移植构建组织工程化皮肤替代物移植后促进了新生血管形成、缩短了皮肤创面的修复。实验显示 GMSCs 作为种子细胞复合 ADM,移植构建组织工程化皮肤替代物可用于皮肤缺损的修复治疗。

목적::탐토아간간충질간세포(GMSCs)복합탈세포진피기질(ADM)수복피부결손적실험。방법:이순화3대 GMSCs 작위충자세포접충우 ADM,구건조직공정화피부체대물。실험분조:18지 SD대서수궤분성3조:GMSCs 복합 ADM 이식조、단순 ADM 조화공백대조조。장3조상응재료분별이식우 SD 대서배부피부결손처,관찰비교창면수복효과。결과:GMSCs 복합 ADM 이식조、단순 ADM 조화공백대조조피부결손창면유합시간분별위(15.1±1.3)d、(18.5±2.1)d 화(22.6±2.6)d,차이유현저성의의(P <0.05)。조직학관찰현시,GMSCs 복합 ADM 이식조성섬유세포、신생소혈관、효원섬유균교단순 ADM 이식조화공백대조조명현증다。결론:GMSCs 복합 ADM 이식구건조직공정화피부체대물이식후촉진료신생혈관형성、축단료피부창면적수복。실험현시 GMSCs 작위충자세포복합 ADM,이식구건조직공정화피부체대물가용우피부결손적수복치료。
Objective:To investigate the feasibility of Gingiva mesenchymal stem cells(GMSCs)as the seeding cells which are inoculated in Acellular Dermal Matrix(ADM)for skin defect repair.Methods:GMSCs of generation 3,the seeding cells,were inoculated in Acellular dermal matrix (ADM)gel to construct the tissue engineered skin substitute.Eighteen SD rats were were randomly divided into three groups experiment group which was treated with a graft about GMSCs and ADM tissue,single ADM and control groups.In order to observe the repair effect on wound,the materials were separately grafted onto SD rat’s back wounds in the rats.Results:The time of wound healing in the GMSCs and ADM experimental group,the single ADM control group and the blank control group were 15.1±1.3,18.5±2.1 and 22.6 ±2.6 days,respectively,and the differences were significant (P < 0.05).Histological observation indicated that the fibroblasts,new vascularities and collagen fibers in the GMSCs and ADM experimental group were obviously increased as compared with those in single ADM group or in control group.Conclusion:GMSCs and ADM can accelerate the formation of fresh bloodvessels of the wounds and promote the skin wound healing.It suggests that GMSCs and ADM maybe an optional method of skin reconstruction for wound repair treatment.
