目的::探讨单侧肾切除联合 STZ 诱导 SD 大鼠糖尿病肾病(DN)造模过程中的注意事项及动物成模后的病理改变。方法:健康雄性大鼠24只,随机分为2组:假手术组(S 组)和糖尿病肾病模型组(DN 组)。DN 组采用左侧肾切除,术后2周腹腔注射 STZ 方法建立大鼠 DN 模型,以造模后72 h 非空腹血糖达到16.8 mmol/L 为模型成立标准。观察 DN 大鼠体重、饮水量、进食量、尿量和血糖变化,于成模后4周处死动物,测定血清肌酐(SCr)、尿素氮(BUN)含量,观察肾组织形态学变化。结果:与 S 组相比, DN 组大鼠于成模后体重增速明显放缓(P <0.01),进食量和饮水量增加,24 h 尿量增多(P <0.01),非空腹血糖维持在成模标准之上。DN 组大鼠 BUN 和 SCr 含量较 S 组大鼠明显升高,肾脏病理切片提示 DN组大鼠肾皮质细胞明显受损。结论:单侧肾切除联合 STZ 诱导可以建立稳定的大鼠 DN 模型。

목적::탐토단측신절제연합 STZ 유도 SD 대서당뇨병신병(DN)조모과정중적주의사항급동물성모후적병리개변。방법:건강웅성대서24지,수궤분위2조:가수술조(S 조)화당뇨병신병모형조(DN 조)。DN 조채용좌측신절제,술후2주복강주사 STZ 방법건립대서 DN 모형,이조모후72 h 비공복혈당체도16.8 mmol/L 위모형성립표준。관찰 DN 대서체중、음수량、진식량、뇨량화혈당변화,우성모후4주처사동물,측정혈청기항(SCr)、뇨소담(BUN)함량,관찰신조직형태학변화。결과:여 S 조상비, DN 조대서우성모후체중증속명현방완(P <0.01),진식량화음수량증가,24 h 뇨량증다(P <0.01),비공복혈당유지재성모표준지상。DN 조대서 BUN 화 SCr 함량교 S 조대서명현승고,신장병리절편제시 DN조대서신피질세포명현수손。결론:단측신절제연합 STZ 유도가이건립은정적대서 DN 모형。
Objective:To investigate the influencing factors in the establishment of experimental model of diabetic nephropathy induced by unilateral nephritic resection and STZ in rats,and to discuss the pathological changes after the animal model was established.Methods:A total of 24 male SD rats were randomly divided into sham (S)group and diabetic nephropathy (DN)group.Diabetic nephropathy model was made by administration of streptozotocin (STZ)in a single dose of 50 mg/kg intraperitoneally 2 weeks after the left kidney was removed. The model was considered successful when the level of non-fasting blood glucose was above 16.8 mmol/L 72 h after STZ injection.Changes of body weight,water intake,food intake,urine volume and blood glucose of DN rats were observed.Rats were sacrificed 4 weeks after model establishment,and renal function was detected by serum creatinine(SCr)and urea nitrogen(BUN)determination.Changes of renal tissue morphology were observed.Results:Compared with S group,the body weight growth of DN group was significantly slowed (P <0.01), while food and water intake increased,24 hours urine volume significantly increased(P<0.01),and the non-fasting blood glucose level higher than that before model establishment.The level of BUN and SCr in DN group was significantly raised than in S group.Renal pathological section showed that renal cortex cells of DN group were significantly impaired.Conclusion:Unilateral nephritic resection and STZ injection is an effective way to induce stable rat model of diabetic nephropathy.
