目的::探讨快速老化小鼠认知功能减退与海马 CA1区锥体细胞树突棘变化的关系。方法:选用6月龄雄性 SAM-P8和 SAM-R1小鼠各8只,分别作为实验组和对照组,通过 Morris 水迷宫实验检测小鼠认知状况,通过 Golgi 染色观察海马 CA1区树突棘的变化。结果:与 SAM-R1小鼠比较,SAM-P8小鼠逃避潜伏期明显延长,跨越平台次数明显减少,海马 CA1区树突棘密度明显减少,差异均具有统计学意义(P <0.01);Morris 水迷宫实验中第5天逃避潜伏期及跨越平台次数与海马 CA1区树突棘密度分别呈负、正相关(P <0.01)。结论:SAM-P8小鼠海马 CA1区树突棘密度的减少导致小鼠认知功能减退,可能与突触功能障碍和突触丢失相关。

목적::탐토쾌속노화소서인지공능감퇴여해마 CA1구추체세포수돌극변화적관계。방법:선용6월령웅성 SAM-P8화 SAM-R1소서각8지,분별작위실험조화대조조,통과 Morris 수미궁실험검측소서인지상황,통과 Golgi 염색관찰해마 CA1구수돌극적변화。결과:여 SAM-R1소서비교,SAM-P8소서도피잠복기명현연장,과월평태차수명현감소,해마 CA1구수돌극밀도명현감소,차이균구유통계학의의(P <0.01);Morris 수미궁실험중제5천도피잠복기급과월평태차수여해마 CA1구수돌극밀도분별정부、정상관(P <0.01)。결론:SAM-P8소서해마 CA1구수돌극밀도적감소도치소서인지공능감퇴,가능여돌촉공능장애화돌촉주실상관。
Objective:To explore the relationship between the decline of cognitive function and the changes of dendritic spines of pyramid cell in hippocampal CA1 region in senescence accelerated mouse.Methods:To select 6-month-old male SAM-P8 and SAM-R1 as experimental group and control group respectively(8 in each group).Cognitive situation of mice in each group were tested by Morris water maze(MWM) test,and the changes of dendritic spines in hippocampal CA1 region were observed by Golgi staining.Results:Compared with SAM-R1,the escape latency was obviously prolonged,the times of crossing platform was obviously decreased.the density of dendritic spines in hippocampal CA1 region was reduced conspicuously in SAM-P8,and all of the differences were statistically significant(P<0.01).The escape latency of the fifth day and the times of crossing platform in MWM test were negativelyand positive respectively correlated to the density of dendritic spines in hippocampal CA1 region(P <0.01).Conclusion:The reduce of the density of dendritic spines in hippocampal CA1 region lead to the decline of cognitive function in SAM-P8,which may be associated with synaptic dysfunction and the loss of synaptic.
