探讨不同处理方式海马蛋白的差异,并对其部分生化特性进行研究。利用硫酸铵分步沉淀、聚丙烯酰胺凝胶电泳法(SDS-PAGE)、bandscan软件、红外图谱对蒸馏水复水,醋酸铵复水,烘烤炮制获得的海马蛋白质进行比较分析。结果发现,烘烤炮制的海马蛋白质结构被破坏,凝胶图谱无条带,而不同复水方法凝胶图谱具有显著差异;通过计算分子量得知海马中蛋白质主要为肌肉蛋白质中的肌浆蛋白与肌原纤维蛋白;对海马蛋白质的红外指纹图谱进行分析,海马不同处理方式提纯后的蛋白质与烘烤炮制的海马全蛋白质的图谱存在区别。

To investigate the difference of hippocampal proteins by difference processing pathways and to study its biochemical properties. Analysis on characteristic from hippocampal protein by difference processing pathways (rehydration by water, rehydration by ammonium acetate, processing by bake) by method of fractional precipitation, SDS-PAGE, bandscan5.0, Infrared spectrometry. It was found that the protein form hippocampus which processing by bake structure was destroyed, there was no gel band pattern. The protein of hippocampal rehydration by water was different from rehydration by ammonium acetate significantly. Calculated molecular weight of the protein in the hippocampus,we can known that the major protein in hippocampus were SPP and MFP. There were some differences between the IR spectra of hippocampus proteins by difference processing pathways.
