目的:比对羊水、血浆蛋白质指纹图谱,寻找羊水特异性表达蛋白,为产科学的临床研究提供分子依据。方法应用表面增强激光解吸电离飞行时间质谱仪采用4种不同原理的蛋白芯片检测了12名正常足月产妇的羊水、血浆蛋白质指纹图谱。结果羊水和血浆标本分别测得蛋白信号(信噪比大于3)150个和250个。4种芯片检出羊水特异性蛋白30个,相对分子质量为11198、12705 Da 的蛋白是比较确定的羊水特异性蛋白。结论羊水中存在特异性表达蛋白,其在妊娠相关生理、病理学中可能起重要作用。

Objective To explore the proteins specifically-expressed in the amniotic fluid (AF)by comparing the fingerprint patterns of in the AF and plasma proteins,aiming to provide molecular evidence for obstetrics research.Methods Surface enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS)with four different protein-chips was utilized to investigate the fingerprint patterns of AF and plasma proteins from 1 2 healthy full-term pregnant women.Results In total,1 50 protein signals (signal-to-noise ratio >3)were detected in the AF and 250 in the plasma.Thirty proteins were detected specifically ex-pressed in the AF using four types of protein-chips.The proteins with a molecular weight of 1 1 1 98 Da and 1 2705 Da were confirmed to be specifically expressed in the AF.Conclusion The AF contains specifically-expressed proteins,which probably play a vital role in the physiological and pathological progresses during pregnancy.
