背景:前期研究表明,疮灵液具有较好的抗炎作用,可抑制血小板聚集及血栓形成,促进伤口愈合。目的:观察将疮灵液载入胶原后对慢性创面愈合的影响。方法:在40只大鼠背部制作慢性创面模型,造模后3 d随机均分为4组,对照组以载0.2 mL生理盐水纱布外敷创面,疮灵液组以载0.2 mL疮灵液纱布外敷创面,胶原组以胶原海绵覆盖创面,疮灵液载入胶原组以载0.2 mL疮灵液胶原覆盖创面。造模时及造模后第3,7,15天测定创面面积,创面分泌物中白细胞总数、白细胞介素6与肿瘤坏死因子α水平,创面肉芽中羟脯氨酸、基质金属蛋白酶1,2,9的水平。结果与结论:疮灵液载入胶原组、疮灵液组造模后第3,7,15天肿瘤坏死因子α、基质金属蛋白酶1,2,9水平均低于对照组、胶原组(P <0.01,P <0.05),造模后第3,7,15天白细胞总数显著低于对照组、胶原组(P <0.01,P <0.05),前两组间各指标比较差异无显著性意义;疮灵液载入胶原组造模后第3,7,15天羟脯氨酸水平高于其余3组(P <0.05);疮灵液载入胶原组造模后15 d创面面积显著低于其余3组(P <0.01);造模后15 d,4组间白细胞介素6水平比较差异无显著性意义。表明疮灵液载入胶原能显著抑制慢性创面炎症细胞增殖与浸润,抑制创面炎症因子与基质金属蛋白酶分泌,促进慢性创面愈合。
揹景:前期研究錶明,瘡靈液具有較好的抗炎作用,可抑製血小闆聚集及血栓形成,促進傷口愈閤。目的:觀察將瘡靈液載入膠原後對慢性創麵愈閤的影響。方法:在40隻大鼠揹部製作慢性創麵模型,造模後3 d隨機均分為4組,對照組以載0.2 mL生理鹽水紗佈外敷創麵,瘡靈液組以載0.2 mL瘡靈液紗佈外敷創麵,膠原組以膠原海綿覆蓋創麵,瘡靈液載入膠原組以載0.2 mL瘡靈液膠原覆蓋創麵。造模時及造模後第3,7,15天測定創麵麵積,創麵分泌物中白細胞總數、白細胞介素6與腫瘤壞死因子α水平,創麵肉芽中羥脯氨痠、基質金屬蛋白酶1,2,9的水平。結果與結論:瘡靈液載入膠原組、瘡靈液組造模後第3,7,15天腫瘤壞死因子α、基質金屬蛋白酶1,2,9水平均低于對照組、膠原組(P <0.01,P <0.05),造模後第3,7,15天白細胞總數顯著低于對照組、膠原組(P <0.01,P <0.05),前兩組間各指標比較差異無顯著性意義;瘡靈液載入膠原組造模後第3,7,15天羥脯氨痠水平高于其餘3組(P <0.05);瘡靈液載入膠原組造模後15 d創麵麵積顯著低于其餘3組(P <0.01);造模後15 d,4組間白細胞介素6水平比較差異無顯著性意義。錶明瘡靈液載入膠原能顯著抑製慢性創麵炎癥細胞增殖與浸潤,抑製創麵炎癥因子與基質金屬蛋白酶分泌,促進慢性創麵愈閤。
배경:전기연구표명,창령액구유교호적항염작용,가억제혈소판취집급혈전형성,촉진상구유합。목적:관찰장창령액재입효원후대만성창면유합적영향。방법:재40지대서배부제작만성창면모형,조모후3 d수궤균분위4조,대조조이재0.2 mL생리염수사포외부창면,창령액조이재0.2 mL창령액사포외부창면,효원조이효원해면복개창면,창령액재입효원조이재0.2 mL창령액효원복개창면。조모시급조모후제3,7,15천측정창면면적,창면분비물중백세포총수、백세포개소6여종류배사인자α수평,창면육아중간포안산、기질금속단백매1,2,9적수평。결과여결론:창령액재입효원조、창령액조조모후제3,7,15천종류배사인자α、기질금속단백매1,2,9수평균저우대조조、효원조(P <0.01,P <0.05),조모후제3,7,15천백세포총수현저저우대조조、효원조(P <0.01,P <0.05),전량조간각지표비교차이무현저성의의;창령액재입효원조조모후제3,7,15천간포안산수평고우기여3조(P <0.05);창령액재입효원조조모후15 d창면면적현저저우기여3조(P <0.01);조모후15 d,4조간백세포개소6수평비교차이무현저성의의。표명창령액재입효원능현저억제만성창면염증세포증식여침윤,억제창면염증인자여기질금속단백매분비,촉진만성창면유합。
BACKGROUND:It has been shown thatChuanglingye has good anti-inflammatory effects, inhibits platelet aggregation and thrombus and improve wound healing. OBJECTIVE:To investigate the effects ofChuanglingye-loaded colagen on regulating inflammation and healing process of chronic wounds. METHODS: Forty rats were selected to make chronic wound models and randomly divided into four groups at 3 days after modeling: control group, colagen group,Chuanglingye group,Chuanglingye-loading colagen group, 10 rats in each group, respectively treated with 0.2 mL normal saline, 0.2 mL Chuanglingye, colagen and Chuanglingye-loaded colagen. The size of wounds was measured when modeling and at 3, 7 and 15 days after modeling. Amount of white blood cels, interleukin-6 level and tumor necrosis factor-α level in wound exudates were detected. The levels of hydroxyproline and matrix mentaloproteases (MMPs), including MMP-1, MMP-2, MMP-9, in the granulation tissue, were also detected. RESULTS AND CONCLUSION:At 3, 7, 15 days after modeling, the amount of white blood cels and levels of interleukin-6, tumor necrosis factor-α, MMP-1, MMP-2, MMP-9 were significantly lower in the Chuanglingye-loaded colagen group andChuanglingye group compared to the control group and colagen group (P < 0.01, P < 0.05). But there was no significant difference betweenChuanglingye-loaded colagen group and <br> Chuanglingye group. The level of hydroxyproline in the granulation tissue was significantly higher in the Chuanglingye-loaded colagen group than the other three groups at 3, 7, 15 days after modeling (P < 0.05), and the size of wound in theChuanglingye-loaded colagen group was significantly lower than that in the other three groups at 15 days after modeling (P < 0.01). In addition, no significant difference was found among the four groups at 15 days after modeling. These findings indicate thatChuanglingye-loaded colagen can significantly depress proliferation and infiltration of inflammatory cels on the wound surface, decrease levels of inflammatory factors and MMPs in wound exudates, and thus promote the healing of chronic wounds.
