研究目的:通过口服补肝肾、续筋骨类中药,观察该药对大鼠膝关节股骨髁间凹软骨全层缺损的修复情况,探讨该类中药对软骨缺损修复的影响.研究方法:选用sD大鼠42只,以手术钻孔的方法,造成左膝股骨髁间全层软骨缺损.实验组每日两次灌服复方中药3ml,对照组无治疗,于术后3周、6周、9周,各处死大鼠7只,取股骨下端,作H.E染色、美蓝染色,观察组织学改变,并作胶原Ⅱ型和肝细胞生长因子(hepatocyte growth factor ,HGF)的免疫组化染色.研究结果:组织学结果显示,9周时实验组组织学修复优于对照组;实验组Ⅱ型胶原表达较对照组有增强的趋势;HGF在软骨细胞胞浆表达,且发现肌肉组织的肌浆中有阳性表达.研究结论:运用中医补肝肾、强筋骨的方法,可以改善局部营养供应,提高代谢机能,从而达到大鼠膝关节软骨缺损修复的目的.

연구목적:통과구복보간신、속근골류중약,관찰해약대대서슬관절고골과간요연골전층결손적수복정황,탐토해류중약대연골결손수복적영향.연구방법:선용sD대서42지,이수술찬공적방법,조성좌슬고골과간전층연골결손.실험조매일량차관복복방중약3ml,대조조무치료,우술후3주、6주、9주,각처사대서7지,취고골하단,작H.E염색、미람염색,관찰조직학개변,병작효원Ⅱ형화간세포생장인자(hepatocyte growth factor ,HGF)적면역조화염색.연구결과:조직학결과현시,9주시실험조조직학수복우우대조조;실험조Ⅱ형효원표체교대조조유증강적추세;HGF재연골세포포장표체,차발현기육조직적기장중유양성표체.연구결론:운용중의보간신、강근골적방법,가이개선국부영양공응,제고대사궤능,종이체도대서슬관절연골결손수복적목적.
Objective To observe the effect ot Chinese herbs in repairing the detect ot articular cartilage ot the knee, and try to discuss its mechanism. Methods The drill, full-thick articular cartilage defects were produced in the left knee joints of 42 rats. Every rat of experimental group was feeding with Chinese herbs 3ml two times a day as treated. 7 animals were killed respectively in 3, 6 and 9 weeks, and the histological observation, H. E pigmentation and immunohistochemical examination were performed. Immunohistochemical staining of type-Ⅱ collagen and hepatocyte growth factor (HGF) were analyzed by image-pro-plus system. Result The repairing tissue in experimental group was better than control group. The Expression of type-Ⅱ collagen increased than control group. HGF expressed in cartilage cell plasm, and showed positive in muscle plasm. Conclusion Chinese herbs is nourishing and enriching the liver and kidney, it plays an important role in repairing defect of articular cartilage of the knee. The effective mechanism is to replenish materials, and strengthen the physiological function.
