观察不同强度有氧耐力训练后的骨骼肌PPARα/δ表达、相关酶活性、肌纤维类型和运动能力的变化,探究PPARα/δ在提高C57BL/6小鼠骨骼肌有氧耐力中的作用以及训练强度的影响。选用6周龄雄性C57BL/6小鼠45只,随机分为3个实验组,分别为76%VO2max 60min训练组(LS)、86%VO2max60min训练组(HS)和安静对照组(RC)。各运动组进行跑台训练,采用RT-PCR、Westernblot、MATPase染色检测骨骼肌中PPARα/δ、CPT-1、BoAcDH和GPDH的表达及肌纤维类型的变化。结果发现:运动各组骨骼肌PPARα/δ转录和翻译水平较对照组均有显著提高,而HS组均显著高于LS组;各运动组CPT-1、BoAc—DH的蛋白袁达和运动能力较对照组均有显著提高,但运动组间没有显著差异,GPDH的表达没有显著改变;运动各组耐力能力较对照组均有显著增强,但运动组间没有显著差异;各组之间的骨骼肌Ⅰ、Ⅱ型肌纤维组成百分比均没有显著差异。结论:PPARα/δ可能作为靶点在骨骼肌对耐力训练的适应过程中发挥着重要作用,高强度训练对于这种适应过程的影响较大;运动并没有导致骨骼肌纤维类型发生显著变化。

Objective:From this view, the expression of PPARα and PPARδ, the activity of some interrelated enzymes (CPT-1, BoAcDH and GPDH), the fiber types in skeletal muscles and the exercise capacity were studied on different exercise intensity of endurance training. Based on these studies, this paper discusses the role of PPARα/δ in skeletal muscle aerobiendurance enhancement of C57BL/6 mice and the effects of Different exercise intensity of endurance training on them. Method: Male C57BL/6 mice (6 weeks old) were randomly divided into three groups (two exercise groups and one control group) : medium and low intensity (12m/min) (LS) ,medium and high intensity (20m/min) (HS), and no training (RC). Every exercise group did treadmill training for six weeks. After each training program, the mice were sacrificed, and tissues were removed. RT-PCR, Western blot and mATPase staining were applied to analyze the effects of endurance training on skeletal muscle PPARα/δmRNA, PPARα/δ. CPT- 1 . BoAcDH 和 GPDH protein expression and their relationship with skeletal muscle types. Result:. The expression of PPARα/δ has an significant increase by Different exercise intensity of Endurance Training in every training group, and HS has an significant increase than LS, And endurance trainings induce the improvement of the enzyme activity and then a higher level of oxidation and metabolism in skeletal muscles, the expression of GPDH has no change, and training locomotory capacity has an significant increase. Aerobic exercise did not cause significant change in muscle fiber type implicate that skeletal muscle type switch may be affected by both cellular metabolism and some other unknown factors. Conclusion.PPARα/δ could play an important role on the adaption of skeletal muscle for endurance training as a target. This adaption is activated more easily under higher intensity. Aerobic exercise did not cause significant change in muscle fiber type.
