目的:探索过度训练对大鼠骨骼肌肌球蛋白重链(MHC)亚型基因表达的影响,为进一步研究其蛋白水平及基因水平的变化机制提供依据。方法:将雄性田大鼠随机分为2组:A、安静对照组(Control,n=10),B、过度训练组(Over-training,OT,n=10)。过度训练组:进行持续性大运动量跑台力竭训练8周,两组均于安静状态下宰杀,迅速选取股外侧肌样本液氮保存。用实时荧光-FCR法,测定骨骼肌MHCⅠ,Ⅱa,Ⅱx,Ⅱb四种亚型的mRNA基因表达。结果:过度训练组MHCⅠmRNA及MHC Ⅱb mRNA明显高于安静对照组(P〈0.05);MHCⅡa mRNA及MHCⅡx mRNA两组与对照无明显差异(P〉0.05)。另一方面,MHC各亚型所占百分比亦有相应的变化,即训练组MHCⅠ和MHCⅡb mRNA百分比较对照组显著增加,而MHCⅡa和MHCⅡx mRNA百分比则略有下降。由于慢Ⅰ型比例很小,从快Ⅱ亚型来看,可见“由快变慢”的变化趋势。提示本运动模型下,各亚型的变化并不一致,具有选择性。这就为下一步研究成肌调节因子的调节机制提供了依据。

목적:탐색과도훈련대대서골격기기구단백중련(MHC)아형기인표체적영향,위진일보연구기단백수평급기인수평적변화궤제제공의거。방법:장웅성전대서수궤분위2조:A、안정대조조(Control,n=10),B、과도훈련조(Over-training,OT,n=10)。과도훈련조:진행지속성대운동량포태력갈훈련8주,량조균우안정상태하재살,신속선취고외측기양본액담보존。용실시형광-FCR법,측정골격기MHCⅠ,Ⅱa,Ⅱx,Ⅱb사충아형적mRNA기인표체。결과:과도훈련조MHCⅠmRNA급MHC Ⅱb mRNA명현고우안정대조조(P〈0.05);MHCⅡa mRNA급MHCⅡx mRNA량조여대조무명현차이(P〉0.05)。령일방면,MHC각아형소점백분비역유상응적변화,즉훈련조MHCⅠ화MHCⅡb mRNA백분비교대조조현저증가,이MHCⅡa화MHCⅡx mRNA백분비칙략유하강。유우만Ⅰ형비례흔소,종쾌Ⅱ아형래간,가견“유쾌변만”적변화추세。제시본운동모형하,각아형적변화병불일치,구유선택성。저취위하일보연구성기조절인자적조절궤제제공료의거。
Objective: To explore the influences of over-training on the gene expression of myosin heavy chain (MHC) isoforms in rats' skeletal muscle, and to supply some basis for further research on its mechanism of level of protein and gene. Method: 20 male SD rats are divided into 2 groups randomly: A. Control, n= 10, B. Over-training, OF, n= 10. The OF Group carries on the heavy-load running exercise to exhaustion for 8 weeks. Then both groups are killed at quiet situation. The vastus lateralis samples are selected and preserved in liquid nitrogen quickly. The mRNA gene expression of the four MHC isoforrns of the skeletal muscle is measured by real-time PCR. Resuhs: The gene expression of MHC isoforms measured by real-time PCR shows that the over-training group is apparendy higher than the quiet control group in MHC Ⅰ mRNA and MHC Ⅱb mRNA (P〈0.05) ; There is no apparent differences between the two groups in MHC Ⅱx mRNA and MHC Ⅱa mRNA (P 〉0.05). Percentage change in gene expression of MHC isoforms at rest after treadmill training is similar. The trend of "from quick to slow" exists. The changes of isoforms are not consistent and are selective on this exercise model. This supplied basis for further research on the regulation mechanism of myogenic regulation factors (MRFs).
