目的:探讨连续离心运动训练对骨骼肌中Calpain和Ubiquitin含量的影响及其与骨骼肌损伤之间的关系;方法:雄性SD大鼠29只随机分为对照组、离心训练后即刻组、24h组和7天组。训练组大鼠连续进行7天的下坡跑运动,分别在末次训练后即刻,24h和第7天取股四头肌,测定股四头肌的超微结构、血清LDH和CK活性以及骨骼肌中Calpain-1、Calpain-2和Ubiquitin含量;结果:1)7天离心训练后即刻和24h,股四头肌超微结构的损伤性变化呈渐进性加重,离心训练后24h组出现明显的肌丝坏死。训练后第7天仍不能完全恢复。血清CK和LDH活性的变化与大鼠股四头肌超微结构的变化相一致。2)与对照组相比,末次训练后即刻骨骼肌中Calpain-1、Calpain-2和Ubiquitin含量均显著升高;训练后24h,Calpain-1和Calpain-2含量进一步升高,其中,Calpain-1显著高于对照组和训练后24h组,Calpain-2虽显著高于对照组,但与训练后即刻组相比无显著性差异;而Ubiquitin含量显著低于训练后即刻组。训练后第7天,Calpain-1和Calpain-2含量显著低于训练后24h组,但仍然高于对照组;而Ubiquitin含量虽高于训练后24h组和对照组,但无显著性差异;结论:1)连续的离心运动可能对骨骼肌纤维的损伤产生一定的累加作用;2)短期连续的离心训练后,骨骼肌中Calpain和Ubiquitin的动态变化几乎与骨骼肌超微结构的动态变化相一致。离心运动训练导致骨骼肌中Calpain和Ubiquitin含量的增加,可能是导致骨骼肌累积性损伤的重要机制。

목적:탐토련속리심운동훈련대골격기중Calpain화Ubiquitin함량적영향급기여골격기손상지간적관계;방법:웅성SD대서29지수궤분위대조조、리심훈련후즉각조、24h조화7천조。훈련조대서련속진행7천적하파포운동,분별재말차훈련후즉각,24h화제7천취고사두기,측정고사두기적초미결구、혈청LDH화CK활성이급골격기중Calpain-1、Calpain-2화Ubiquitin함량;결과:1)7천리심훈련후즉각화24h,고사두기초미결구적손상성변화정점진성가중,리심훈련후24h조출현명현적기사배사。훈련후제7천잉불능완전회복。혈청CK화LDH활성적변화여대서고사두기초미결구적변화상일치。2)여대조조상비,말차훈련후즉각골격기중Calpain-1、Calpain-2화Ubiquitin함량균현저승고;훈련후24h,Calpain-1화Calpain-2함량진일보승고,기중,Calpain-1현저고우대조조화훈련후24h조,Calpain-2수현저고우대조조,단여훈련후즉각조상비무현저성차이;이Ubiquitin함량현저저우훈련후즉각조。훈련후제7천,Calpain-1화Calpain-2함량현저저우훈련후24h조,단잉연고우대조조;이Ubiquitin함량수고우훈련후24h조화대조조,단무현저성차이;결론:1)련속적리심운동가능대골격기섬유적손상산생일정적루가작용;2)단기련속적리심훈련후,골격기중Calpain화Ubiquitin적동태변화궤호여골격기초미결구적동태변화상일치。리심운동훈련도치골격기중Calpain화Ubiquitin함량적증가,가능시도치골격기루적성손상적중요궤제。
Objective:To investigate the effects of continuous eccentric exercise training on Calpain and Ubiquitin content skeletal muscle and its relationship with skeletal muscle injury.Methods:29 male SD rats were randomly divided into control group,immediately post-training group(ET0),24h post-training group(ET24)and 7 days post-training group(ET7d).Training rats consecutive downhill running for 7 days.Quadriceps were taken immediately,24h and 7th day after the last bout of eccentric exercise respectively,the changes of ultrastructure,serum LDH and CK activity and Calpain-1,calpain-2,Ubiquitin concentrations were observed.Results:1)Ultrastructural changes of quadriceps injury showed gradual increase immediately and 24h at the last bout of eccentric exercise,the apparent necrosis of myofilament occurred at 24h post-training.It still not fully recovered at 7th day after training.The changes of serum CK and LDH activity had consistent with ultrastructural changes.2)Compared with the control group,Calpain-1,Calpain-2 and Ubiquitin levels in skeletal muscle were significantly increased immediately after the last training.At 24h post-training,Calpain-1 and Calpain-2 levels further increased,including Calpain-1 significantly higher than that in control group and ET0 group,Calpain-2 significantly higher than in control group,but there was no significant difference compared with ET0 group,while Ubiquitin content was significantly lower than that in ET0 group.At the 7th post-training,Caipain-1 and Calpain-2 were significantly lower than those in ET24 group,but still higher than control group.While Ubiquitin levels in ET7d group was higher than that in ET24 group and the control group,but there were no significant difference.Conclusion:1)Repeated eccentric exercise might produce a certain cumulative effect on skeletal muscle damage.2)The dynamic changes of Calpain and Ubiquitin content in skeletal muscle after short-term repeated eccentric training was almost consistent with the dynamic changes of skeletal muscle ultrastructure.Eccentric exercise training might result in the increase of Calpain and Ubiquitin levels in skeletal muscle,which might be an important mechanism about cumulative damage of skeletal muscle.
