目的:观察补充白藜芦醇对运动引起的骨骼肌线粒体形态学变化及线粒体Ca2+变化的影响。方法:70只雄性昆明种小鼠随机分为安静对照组(control group,CG)、运动对照组(exercise group,EG)和运动+白藜芦醇组(exercise+resveratrol group,ER),EG组、ER组又分为3个亚组,运动后即刻组、运动后24 h组、运动后48 h组,每组10只。ER组每日给予白藜芦醇10 mg/kg灌胃,与EG组同时进行4周的跑台训练,训练结束后进行一次坡度为0,速度为18 m/min,持续90 min的跑台运动。运动结束后按组别取材,电镜观察肌骨骼肌线粒体形态学变化;测定肌组织SOD活性、MDA含量、线粒体Ca2+浓度。结果:电镜结果表明,EG组小鼠运动后骨骼肌线粒体出现了肿胀、空化、髓样化等现象;ER组小鼠运动后骨骼肌线粒体虽也出现相似病变,但在运动后48 h线粒体病变明显减轻;ER组骨骼肌中SOD活性显著升高、MDA含明显降低,与EG组相比,差异具有显著性(P〈0.05);EG组运动后各时刻线粒体Ca2+含量明显增加,48 h后仍末恢复(P〈0.05);ER组运动后虽线粒体Ca2+含量也明显增加,但运动后48 h已明显降低,差异具有显著性(P〈0.05)。结论:一次长时间剧烈运动可引起骨骼肌线粒体发生损伤性变化;补充白藜芦醇可以提高骨骼肌抗氧化能力,降低脂质过氧化反应,调节线粒体Ca2+变化,对线粒体的结构与功能具有一定的保护作用。

목적:관찰보충백려호순대운동인기적골격기선립체형태학변화급선립체Ca2+변화적영향。방법:70지웅성곤명충소서수궤분위안정대조조(control group,CG)、운동대조조(exercise group,EG)화운동+백려호순조(exercise+resveratrol group,ER),EG조、ER조우분위3개아조,운동후즉각조、운동후24 h조、운동후48 h조,매조10지。ER조매일급여백려호순10 mg/kg관위,여EG조동시진행4주적포태훈련,훈련결속후진행일차파도위0,속도위18 m/min,지속90 min적포태운동。운동결속후안조별취재,전경관찰기골격기선립체형태학변화;측정기조직SOD활성、MDA함량、선립체Ca2+농도。결과:전경결과표명,EG조소서운동후골격기선립체출현료종창、공화、수양화등현상;ER조소서운동후골격기선립체수야출현상사병변,단재운동후48 h선립체병변명현감경;ER조골격기중SOD활성현저승고、MDA함명현강저,여EG조상비,차이구유현저성(P〈0.05);EG조운동후각시각선립체Ca2+함량명현증가,48 h후잉말회복(P〈0.05);ER조운동후수선립체Ca2+함량야명현증가,단운동후48 h이명현강저,차이구유현저성(P〈0.05)。결론:일차장시간극렬운동가인기골격기선립체발생손상성변화;보충백려호순가이제고골격기항양화능력,강저지질과양화반응,조절선립체Ca2+변화,대선립체적결구여공능구유일정적보호작용。
Objective:To observe the effect of resveratrol on exercise-induced skeletal muscle mitochondrial morphology and mitochondrial Ca2+ change.Methods:70 male Kunming mice were randomly divided into three groups:control group(CG),exercise group(EG)and exercise+resveratrol group(ER),and EG group and ER group Were divided into three sub-groups by post-exercise 0 hour,24 hours,48 hours,which every group has 10 mice.The mice of group ER were given resveratrol with 10 mg(/kg·d),and taken protocol of treadmill training for 4 weeks,then had a gradient of 0 and a speed of 18m/ min,continued 90 min of treadmill exercise as did the mice of group EG.Mice were slay respectively.The histological change of mitochondrial were observed by electron microscope.The activity of muscle SOD and MDA,concentration of muscle mitochondria Ca2+ were measured using Ultraviolet Spectrometer.Results:The electron microscope results showed that EG appeared swelling,cavitation,myeloid and some of the mitochondria were irregularly arranged,ER mice Skeletal muscle mitochondria with ridges,membrane fusion,fuzzy unclear after exercise,and the injury level of ER was less than EG.skeletal muscle of ER group SOD activity was significantly increased,MDA with significantly decrease compared with the EG group,a significant difference(P0.05).Compare with CG,the concentration of mitochondria Ca2+ was significantly increased in EG,especially post-exercise 24 h(P0.05).The concentration of mitochondria Ca2+ of ER was significantly lower than that of EG(P0.05).Conclusion:After the long time treadmill exercise,the muscle mitochondrial presents the obvious damage change,supplement resveratrol can improve mice antioxidant capacity,lower lipid peroxidation,and regulation of muscle mitochondrial Ca2+ concentration,so resveratrol could have a certain protective effect on structure and function of mitochondria.
