目的:通过测试萎缩骨骼肌细胞Caspase-3、SOD活性和MDA含量,观察补充扇贝肽对运动大鼠停训导致骨骼肌萎缩后肌细胞Caspase-3活性及其抗氧化能力的影响,研究扇贝肽对停训后骨骼肌萎缩的干预作用。方法:40只SD雄性大鼠以15m/min的速度,持续性水平跑台训练30min开始进行递增负荷运动,运动6d/周,休息1d/周,速度递增3m/min/周,时间递增30min/周,运动3周后即刻处死10只大鼠为基础对照组,其余大鼠随机分组为补充安慰剂阴性对照组、补充牛乳阳性对照组和补充扇贝肽实验组,30只大鼠左侧后肢实施石膏固定模拟停训模型,并进行扇贝肽补充实验。实验组和对照组所有动物每天分别进行灌胃15%扇贝肽饮料2mL、等量的安慰剂和牛乳。结果:发现补充扇贝肽可显著减低停训后萎缩骨骼肌细胞MDA含量增加,增加SOD活性,提高细胞内抗氧化能力,可显著抑制停训后萎缩骨骼肌细胞Caspase-3的活性,从而抑制骨骼肌收缩蛋白和结构蛋白的降解,促进萎缩骨骼肌的恢复。
目的:通過測試萎縮骨骼肌細胞Caspase-3、SOD活性和MDA含量,觀察補充扇貝肽對運動大鼠停訓導緻骨骼肌萎縮後肌細胞Caspase-3活性及其抗氧化能力的影響,研究扇貝肽對停訓後骨骼肌萎縮的榦預作用。方法:40隻SD雄性大鼠以15m/min的速度,持續性水平跑檯訓練30min開始進行遞增負荷運動,運動6d/週,休息1d/週,速度遞增3m/min/週,時間遞增30min/週,運動3週後即刻處死10隻大鼠為基礎對照組,其餘大鼠隨機分組為補充安慰劑陰性對照組、補充牛乳暘性對照組和補充扇貝肽實驗組,30隻大鼠左側後肢實施石膏固定模擬停訓模型,併進行扇貝肽補充實驗。實驗組和對照組所有動物每天分彆進行灌胃15%扇貝肽飲料2mL、等量的安慰劑和牛乳。結果:髮現補充扇貝肽可顯著減低停訓後萎縮骨骼肌細胞MDA含量增加,增加SOD活性,提高細胞內抗氧化能力,可顯著抑製停訓後萎縮骨骼肌細胞Caspase-3的活性,從而抑製骨骼肌收縮蛋白和結構蛋白的降解,促進萎縮骨骼肌的恢複。
목적:통과측시위축골격기세포Caspase-3、SOD활성화MDA함량,관찰보충선패태대운동대서정훈도치골격기위축후기세포Caspase-3활성급기항양화능력적영향,연구선패태대정훈후골격기위축적간예작용。방법:40지SD웅성대서이15m/min적속도,지속성수평포태훈련30min개시진행체증부하운동,운동6d/주,휴식1d/주,속도체증3m/min/주,시간체증30min/주,운동3주후즉각처사10지대서위기출대조조,기여대서수궤분조위보충안위제음성대조조、보충우유양성대조조화보충선패태실험조,30지대서좌측후지실시석고고정모의정훈모형,병진행선패태보충실험。실험조화대조조소유동물매천분별진행관위15%선패태음료2mL、등량적안위제화우유。결과:발현보충선패태가현저감저정훈후위축골격기세포MDA함량증가,증가SOD활성,제고세포내항양화능력,가현저억제정훈후위축골격기세포Caspase-3적활성,종이억제골격기수축단백화결구단백적강해,촉진위축골격기적회복。
Objective: The effects of scallop peptides supplementation on the activity of caspase -3 and SOD and the content of MDA in skeletal muscle after detraining were investigated. To study the effects of scallop peptides supplementation on muscular atrophy after detraining in rats. Methods : 40 male Sprague -Dawley rats ran 30 minutes at speed of 15 m/min , exercise 5 day/week and rest 1 day/week in the first week. In the second week, rats ran 60 minutes at speed of 18 m/min, exercise 5 day/week and rest 1 day/week and ran 90 minutes at speed of 21 m/min, exercise 5 day/week and rest 1 day/week in the third week. After sports training for 3 weeks, Ten rats were killed as baseline group. The 30 rats were randomly divided into 3 groups as placebo, milk and SPs supplemented, Then the left hind limbs of the rats were fixed by plastic gypsum for 3 weeks. Rats in experimental groups were fed 2ml beverage containing 15% SPs, and rats in control were fed 2ml placebo and milk respectively. Results: The results showed that After SPs supplementation, the activity of caspase -3 decreased obviously, and the antioxidant capacity increased obviously in atrophic muscle in raes.
