目的:探讨运动干预对内源性二甲基精氨酸二甲胺水解酶(DDAH)活性及表达的影响。方法:3月龄雌性SD大鼠110只,切除双侧卵巢制造骨质疏松模型。6周后,每组随机选2只大鼠处死,取胫骨上段骨组织,观察模型复制情况。模型复制成后,随机分为5组:对照组,骨质疏松组,低、中、高运动组(游泳)组,正常饲养8周。饲养14周后处死,采集股骨和胫骨组织,经匀浆得酶匀浆。在酶匀浆中的DDAH与ADMA反应,比色测定DDAH的酶活性;用SDS-聚丙烯烯胺凝胶电泳及Western blotting印迹分析法测定二甲基精氨酸二甲胺水解酶表达。结果:骨质疏松组大鼠骨组织的NO、NOS含量均低于对照组和运动组。对照组及3个运动组的大鼠骨组织DDAH的酶活性高于骨质疏松组;不同运动量实验组的骨组织NO、NOS含量和DDAH的酶活性之间无统计学差异。结论:运动会增加骨组织的NO、NOS含量,但并不会影响DDAH的表达;骨质疏松的发生与DDAH的活性下降有关。

목적:탐토운동간예대내원성이갑기정안산이갑알수해매(DDAH)활성급표체적영향。방법:3월령자성SD대서110지,절제쌍측란소제조골질소송모형。6주후,매조수궤선2지대서처사,취경골상단골조직,관찰모형복제정황。모형복제성후,수궤분위5조:대조조,골질소송조,저、중、고운동조(유영)조,정상사양8주。사양14주후처사,채집고골화경골조직,경균장득매균장。재매균장중적DDAH여ADMA반응,비색측정DDAH적매활성;용SDS-취병희희알응효전영급Western blotting인적분석법측정이갑기정안산이갑알수해매표체。결과:골질소송조대서골조직적NO、NOS함량균저우대조조화운동조。대조조급3개운동조적대서골조직DDAH적매활성고우골질소송조;불동운동량실험조적골조직NO、NOS함량화DDAH적매활성지간무통계학차이。결론:운동회증가골조직적NO、NOS함량,단병불회영향DDAH적표체;골질소송적발생여DDAH적활성하강유관。
Objective:To investigate the effects of sports on the activity of osteoporosis DDAH hydrolyzed - enzyme. Method : One hundred and ten female rats of three month' s old were selected as the subjects of the experiment ,which were local anesthesia at the abdomen cavity. Six weeks later, bone tissue of femur and tibia were isolated from the rats to observe the situation of the osteoporosis model. Then the model subjects were randomly divided into five groups: Control Group, Osteoporosis Group and Low- lever Sport Group, Mid -lever Sport Group and High -lever Sport Group. From then on, they were fed normally for 8 weeks and then killed 14 weeks later. Bone tissue of Femur and Tibia were isolated and made into liquid form. The expression of NO and NOS was observed as well as the activity of DDAH, and the DDAH enzyme activity would be illustrated by Western - blot. Result:The NO and NOS from Osteoporosis Group were lower than the other Groups as well as the activity of Osteoporosis DDAH Hydrolyzed - enzyme. However, there is no significant difference between Osteoporosis Group and other there different lever Sport Groups in expression of NO and NOS and the activity of DDAH Hydrolyzed - enzyme. Conclusion : Sports could keep the expression of NO and NOS in normal lever in Osteoporosis and there is a close relationship between the activity of DDAH and Osteoporosis.
