目的:通过对4周负重游泳运动前、后小鼠脾脏CD4+CD25+ Treg表达水平变化及相关细胞因子IL-2、IL-10和TGF—β1水平变化的分析,探讨和研究有氧耐力运动对小鼠脾脏CD4+CD25+Treg表达及细胞因子IL-2、IL-10和TGF-13的影响。方法:KM种雄性小鼠100只随机分为对照组(50只)和运动组(50只)。运动各组分别按运动方案进行45min的负重(5%体重)游泳运动每天1次,每周6次。分别于实验前、每周末次运动后24h,处死小鼠,摘取脾脏和胸腺、采集血样。计算脾指数和胸腺指数,流式细胞仪检测小鼠脾脏CD4+CD25+Treg的表达,ELISA法测定血清IL-2、IL-10和TGF-β含量。结果:在4周负重游泳运动过程中,运动组小鼠脾指数在第1周末增加并在随后2、3、〈周表现出下降的趋势,胸腺指数表现为迈周减少。运动组小鼠脾脏CD4+CD25+Treg的表达率表现为逐周增高,运动3周组、4周组高于运动0周组和对照组(P〈0.05)。运动2周纽和3周组血清IL-2水平升高,高于对照组(P〈0.05);运动3周组和4周组血清IL-10和TGF-β1水平高于对照组(P〈O.05)。结论:4周有氧耐力游泳运动可延缓小鼠脾脏生长,加快胸腺萎缩,进而影响免疫器官的发育和功能。4周有氧耐力游泳运动可诱导小鼠T细胞活化,脾脏CD4+CD25+Treg表达水平提高,血清IL-10和TGF—β1水平升高,诱导免疫偏移。

목적:통과대4주부중유영운동전、후소서비장CD4+CD25+ Treg표체수평변화급상관세포인자IL-2、IL-10화TGF—β1수평변화적분석,탐토화연구유양내력운동대소서비장CD4+CD25+Treg표체급세포인자IL-2、IL-10화TGF-13적영향。방법:KM충웅성소서100지수궤분위대조조(50지)화운동조(50지)。운동각조분별안운동방안진행45min적부중(5%체중)유영운동매천1차,매주6차。분별우실험전、매주말차운동후24h,처사소서,적취비장화흉선、채집혈양。계산비지수화흉선지수,류식세포의검측소서비장CD4+CD25+Treg적표체,ELISA법측정혈청IL-2、IL-10화TGF-β함량。결과:재4주부중유영운동과정중,운동조소서비지수재제1주말증가병재수후2、3、〈주표현출하강적추세,흉선지수표현위매주감소。운동조소서비장CD4+CD25+Treg적표체솔표현위축주증고,운동3주조、4주조고우운동0주조화대조조(P〈0.05)。운동2주뉴화3주조혈청IL-2수평승고,고우대조조(P〈0.05);운동3주조화4주조혈청IL-10화TGF-β1수평고우대조조(P〈O.05)。결론:4주유양내력유영운동가연완소서비장생장,가쾌흉선위축,진이영향면역기관적발육화공능。4주유양내력유영운동가유도소서T세포활화,비장CD4+CD25+Treg표체수평제고,혈청IL-10화TGF—β1수평승고,유도면역편이。
Objective.. By analyzing the changes of CD4+ CD25+ Treg level and related cytokines during four-week load-bearing swimming exercise in mice, the effect of aerobic endurance exer- cise on the CD4+ CD25+ Treg and related cytokines were discus,;ed. Methods.. 100 KM mice were divided into EXE group and CON group. Mice in EXE group swam 45 min per day with a load of 5 % of body weight, and swam 6 day and rested 1 day per week. Mice were executed before the exercise protocol or 24 h after the last swim every week, and spleen and thymus were taken out. Spleen index and thymus index were calculated, Flow Cytometry were used to measure CD4+ CD25+ Treg level, and ELISA were used to determine the levels of IL-2, IL-10 and TGF-β. Results. Spleen index increased firstly and then decreased and thymus index de- creased with swimming going on in EXE group. CD4+ CD25+ Treg level in spleen increased compared with CON group(P〈0.05), and IL-10 and TGF-β evels in EXE 3 week group and four-week group increased compare with CON group(P〈0.05). Conclusion.. Four-week aero- bic endurance exercise delayed spleen growth, promoted thymus atrophy and affected the devel- opment and function of immune organ. Four-week aerobic endurance exercise induced the acti- vation of T cell, expanded CD4+ CD25+ Treg, increased the level cf IL-10 and TGF-β, and led to immunity drift.
