目的:研究运动对肥胖大鼠脂肪组织visfatin基因表达及ACC活性的影响。方法:健康雄性SD大鼠60只(60—95g),随机抽取10只作为普通膳食对照组(C),喂养标准普通饲料。其余50只喂养高脂膳食,7周后建立肥胖大鼠模型16只,再随机分为2组:肥胖非运动组(Ob,n=8)和肥胖运动组(Ob—E,n=8)。8周后测定大鼠体重、血脂、腹内脂肪组织visfatinmRNA表达量及蛋白水平、磷酸化ACC(P—ACC)蛋白水平。结果:大鼠腹内脂肪量Ob组显著高于C组和Ob—E组(P〈0.01或P〈0.05);Ob—E组血清TG明显降低而HDL—C明显升高(P〈0.05);Ob—E组腹内脂肪组织visfatin mRNA表达水平显著低于Ob组(P〈0.05),P—ACC水平显著高于Ob组和C组(P〈0.05);大鼠腹内脂肪组织visfatin蛋白水平没有显著性差异(P〉0.05)。结论:运动降低肥胖大鼠血清TG,提高HDL—C水平,从而改善血脂水平;运动引起肥胖大鼠腹内脂肪量下降,可能与脂肪组织visfatinmRNA表达水平降低和磷酸化ACC水平升高有关;运动对腹内脂肪组织的visfatin蛋白表达水平无显著性影响。

목적:연구운동대비반대서지방조직visfatin기인표체급ACC활성적영향。방법:건강웅성SD대서60지(60—95g),수궤추취10지작위보통선식대조조(C),위양표준보통사료。기여50지위양고지선식,7주후건립비반대서모형16지,재수궤분위2조:비반비운동조(Ob,n=8)화비반운동조(Ob—E,n=8)。8주후측정대서체중、혈지、복내지방조직visfatinmRNA표체량급단백수평、린산화ACC(P—ACC)단백수평。결과:대서복내지방량Ob조현저고우C조화Ob—E조(P〈0.01혹P〈0.05);Ob—E조혈청TG명현강저이HDL—C명현승고(P〈0.05);Ob—E조복내지방조직visfatin mRNA표체수평현저저우Ob조(P〈0.05),P—ACC수평현저고우Ob조화C조(P〈0.05);대서복내지방조직visfatin단백수평몰유현저성차이(P〉0.05)。결론:운동강저비반대서혈청TG,제고HDL—C수평,종이개선혈지수평;운동인기비반대서복내지방량하강,가능여지방조직visfatinmRNA표체수평강저화린산화ACC수평승고유관;운동대복내지방조직적visfatin단백표체수평무현저성영향。
To investigate the effects of exercise on the expression of visfatin gene and the activity of ACC in adipose tissue in obesity rats, 60 healthy male Sprague-Dawley rats(60-95 g) were randomly selected with 10 rats as normal dietary control group of rats (C, n=10) and fed with normal standard diet. The remaining 50 rats were fed high-fat diet. Seven weeks later, 16 diet-induced-obesity rat models were established. They were randomly divided into two group: no-exercise group (Ob, n=8) and exercise group (Ob-E, n=8). Eight weeks later, the body weight and serum lipid of rats, the visfatin mRNA expression and protein levels as well as phosphorylated Acetyl-Coenzyme A carboxylase(P-ACC)levels in visceral adipose tissue of rats were measured. The visceral fat deposit of obesity rats was significantly higher than that in C group and in Ob-E group(P〈0.05). The serum TG in Ob-E group sig- nificantly decreased and the serum HDL-c significantly increased (P〈0.05) ; visfatin mRNA expression levels in visceral adipose tissue of Ob-E group was significantly lower than that of Ob group(P〈0.05) ; The P-ACC levels in visceral adipose tissue of Ob-E group was significantly higher than that of Ob group (P〈0.05) ; visfatin protein level in visceral adipose tissue had no difference among three groups(P〉0.05). Exercise decreased obesity rat's serum TG and increased HDL-c as well as improved serum lipid levels. The visceral fat deposit of obesity rat was decreased by exercise. It might be involved in the decrease of visceral adipose tissue visfatin mRNA expression levels and the increase of P-ACC levels. Exercise did not significantly influence visfatin protein expression levels in visceral adipose tissue.
