目的:探究孕期低强度游泳运动对哺乳期过度喂养子代肥胖大鼠(OF大鼠)下丘脑弓状核食欲调控肽异常表达水平的影响,为预防围产期过度喂养型肥胖提供理论依据。方法:80只SD雌性大鼠妊娠后第3天,随机选取22只母鼠孕期低强度游泳运动至分娩结束(10min/day,6days/week)。母鼠分娩后第2天随机将每窝仔鼠喂养数目调整为4只和10只,断乳后最终获得实验分组为孕期安静+哺乳期正常喂养模型对照组(DBC1,n=8)、孕期安静+哺乳期过度喂养断乳后OF对照组(CSC1,n=8)和孕期低强度游泳运动+哺乳期过度喂养断乳后OF组(ESC1,n=8)。禁食后次日取材,采用原位杂交技术检测下丘脑弓状核NPY、POMC和LRbmRNA表达;放射免疫法测血清胰岛素和瘦素;酶化学法检测血清血糖和血脂。结果:KSCl组的体重、Lee指数、血瘦素、血胰岛素、血糖、IGR、‘TG和TC与cscl组相比无显著降低(P〉0.05);CNC1组下丘脑弓状核POMC和NPYmRNA水平均显著低于CSC1组(Pd0.01),而CNC1组下丘脑弓状核LRbmRNA水平显著高于CSCl组(P〈0.01);ESCl组下丘脑弓状核POMCmRNA显著高于CSCl组(P〈0.01),NPY和U地mRNA水平与CSC1组相比无显著性差异(P〉0.05)。结论:孕期低强度游泳运动和哺乳期过度喂养均可对子代大鼠下丘脑弓状核POMCmRNA表达产生可塑性的影响,但是孕期低强度游泳运动对OF大鼠过度进食诱发的肥胖表型无显著预防效果。

Objective.. To investigate whether the low intensity swimming exercise during preg- nancy could influence the regulation center of appetite and energy in the hypothalamic arcuate nucleus (ARH) of their overfed weanling rats. Methods: Total 80 female SD pregnant rats were enrolled, among which 22 rats began swimming (10 min per day, 6 days per week)from the 3rd day of pregnancy to delivery. The rest rats kept sedentary till delivery. The female pups were distributed randomly and assigned to small litters (4 pups) and normal litters (10 pups) on the second day after birth. After weanling the offspring pups were randomly selected and assigned to three groups according to the body weight., the pregnancy sedentary+normal little control group(CNC1, n=8), the pregnancy sedentary+ postweanling OF control group (CSC1 ,n=8)and the pregnancy swimming + postweanling OF group(ESC1, n=8). The rat pups brains were removed, and NPY, POMC and LRb rnRNA in ARH were analyzed by in situ hybridization technique. The fasting serum insulin and leptin were assayed by radioimmu- noassay, and the fasting serum glucose and lipids were assayed by enzymatic methods. Results : The body weight, Lee index, leptin, insulin, glu, IGR, TC and TG in their pups on the day of weanling, relative to Group CSC1, were not significantly affected in Group ESC1. POMC and NPY rnRNA in ARC exhibitions were significantly lower in Group CNC1 than those of Group CSC1 (P〈0. 01), while LRb mRNA in ARC in Group CNC1 was higher significantly than Group CSC1 (P〉0. 01 ). There were not significant differences between ESC1 and CSC1 groups in the mRNA levels of NPY and LRb in ARH. (P〉0.05), however, POMC mRNA in ARH in Group ESC1 was higher than that of Group GSC1 (P〉0.05). Conclusion.. Both Low-intensity swimming during pregnancy and overfed during lactation have plastic influences in the mRNA expression of POMC in ARH. However low-intensity swimming during pregnan- cy has little significant preventive effect on the weight programming in the overfed rats.
