自噬是一种溶酶体降解通路,是一种在正常和病态细胞中普遍存在的生理机制,对于维持细胞内蛋白质平衡,清除细胞内受损的细胞器及维持内环境稳定起到关键作用。自噬在调节细胞生存和死亡过程中具有双向调节性,并且广泛参与多种生理和病理过程。尽管自噬在癌症、心血管疾病、帕金森病和糖尿病的机理与治疗方面的研究取得了一定进展,而自噬在骨骼肌质量维持中的作用及调控仍不清楚。将自噬在骨骼肌质量维持中功能状态与信号通路做一简要总结,旨在为自噬在骨骼肌中的调控与骨骼肌萎缩提供一个更好的理解,进一步探索骨骼肌萎缩的机制,为预防和治疗肌萎缩提供有效的靶标;同时,适当的运动有利于老年性肌萎缩等老年性疾病的预防与治疗,但细胞自噬的功能状态有着决定性的作用。

Autophagy is a highly conserved lysosomal degradation pathway in mammals as a ubiquitous physiologi- cal mechanism in normal and abnormal cells. This self-eating mode exerts a critical role in maintaining cellular ho- meostasis through degrading excessively accumulated denature proteins and impaired organelles into small mole- cules to provide the blocks for cellular regeneration. Due to its dual function in cell death and cell survival, it is widely involved in many physiological and pathological processes. Although autophagy is extensively explored in pathological and treatment mechanisms of cancer, diabetes, cardiovascular diseases and Parkinson diseases with great achievements, its regulation and role in the mass maintenance of skeletal muscle is still unclear. In this arti- cle, the pathways and functional status of autophagy in maintaining skeletal muscle mass are summarized in order to provide an understanding of the correlation between mechanisms of muscle wasting and effective targets or strategies of prevention and treatments for muscle wasting.
