目的:研究补充烟酸及耐力训练对急性酒精性肝损伤大鼠SifT1表达、活性的影响及机制。研究方法:以Wistar大鼠建立急性酒精性肝损伤模型,以跑台运动为训练手段,并且给予大鼠补充烟酸,测定血清AIJT和As以及肝脏MnSOD活性、NAD+/NADH比值、MDA含量、sirT1活性、线粒体ATP合成活力、MnSOD和sirT1mRNA表达。结果:急性酒精摄入导致血清ALT和AST水平、肝脏MDA含量均显著升高,线粒体ATP合成活力、肝脏NAD’/NADH比值、MnSOD及SirTl活性及mRNA表达均显著降低;耐力训练及补充烟酸再给予酒精摄入与只给予酒精摄入血清ALT和AST水平、肝脏MDA含量、线粒体ArrP合成活力、肝脏NAD+/NADH比值、MnSOD及SirT1活性及mRNA表达变化规律一致,但变化幅度相对较小。结论:耐力训练及补充烟酸均能提高肝脏细胞内NAD+/NADH的比值,使SirT1活性及表达升高,进而提高肝脏细胞抗氧化能力,达到预防/改善急性酒精性肝损伤的目的。

목적:연구보충연산급내력훈련대급성주정성간손상대서SifT1표체、활성적영향급궤제。연구방법:이Wistar대서건립급성주정성간손상모형,이포태운동위훈련수단,병차급여대서보충연산,측정혈청AIJT화As이급간장MnSOD활성、NAD+/NADH비치、MDA함량、sirT1활성、선립체ATP합성활력、MnSOD화sirT1mRNA표체。결과:급성주정섭입도치혈청ALT화AST수평、간장MDA함량균현저승고,선립체ATP합성활력、간장NAD’/NADH비치、MnSOD급SirTl활성급mRNA표체균현저강저;내력훈련급보충연산재급여주정섭입여지급여주정섭입혈청ALT화AST수평、간장MDA함량、선립체ArrP합성활력、간장NAD+/NADH비치、MnSOD급SirT1활성급mRNA표체변화규률일치,단변화폭도상대교소。결론:내력훈련급보충연산균능제고간장세포내NAD+/NADH적비치,사SirT1활성급표체승고,진이제고간장세포항양화능력,체도예방/개선급성주정성간손상적목적。
Objective:To investigate the influence of niacin supplement and endurance training on the activity and expression of SirT1 and its mechanism in alcohol - induced acute hepatic injury in rats. Methods:It analyzed the level of ALT and AST in serum, NAD +/NADH, content of MDA, mito- chondrial ATP synthesis, activities and expressions of MnSOD and SirT1 in liver tissue in alcohol - induced acute hepatic injury rats after treadmill training and niacin supplement. Results: The acute alcohol treatment increases ALT and AST in serum and content of MDA in liver tissue, decreases mitochondrial ATP synthesis, ratio of NAD ~/NADH, activities and expression of MnSOD and SirT1 in alcohol - induced acute hepatic injury rats. The parameters of the above indicates the same after endurance training and niacin supplement but the extent is relatively lower. Conclusion:Endurance training and niacin supplement can increase ration of NAD +/NADH, which improves the activity and expression of SirT1, and then increases antioxidant ability of liver, which achieves the goal of protecting or improving alcoholinduced acute hepatic injury.
