摘要:以河南宋河白酒发酵30d的糟醅为实验材料,通过富集培养及稀释涂布法分离厌氧及兼性厌氧菌,以菌落计数法确定最佳样品稀释度和适宜的厌氧菌培养方法,并采用形态学和生理生化特性进行种属的初步鉴定.结果表明:厌氧及兼性厌氧菌分离适宜稀释度为10^-3,最佳培养方法为抽真空培养法,共分离到5株己酸菌、6株丁酸菌、2株甲烷菌、6株乳酸菌.丁酸菌、己酸菌和乳酸菌革兰氏染色均为阳性,甲烷菌为革兰氏阴性菌.丁酸菌和己酸菌分别归梭菌属(Clostridium)和芽孢杆菌属(Bacillus),甲烷菌为假单胞菌属(Pseudomonas),乳酸菌为芽孢乳杆菌属(Sporolactobacillus).
摘要:以河南宋河白酒髮酵30d的糟醅為實驗材料,通過富集培養及稀釋塗佈法分離厭氧及兼性厭氧菌,以菌落計數法確定最佳樣品稀釋度和適宜的厭氧菌培養方法,併採用形態學和生理生化特性進行種屬的初步鑒定.結果錶明:厭氧及兼性厭氧菌分離適宜稀釋度為10^-3,最佳培養方法為抽真空培養法,共分離到5株己痠菌、6株丁痠菌、2株甲烷菌、6株乳痠菌.丁痠菌、己痠菌和乳痠菌革蘭氏染色均為暘性,甲烷菌為革蘭氏陰性菌.丁痠菌和己痠菌分彆歸梭菌屬(Clostridium)和芽孢桿菌屬(Bacillus),甲烷菌為假單胞菌屬(Pseudomonas),乳痠菌為芽孢乳桿菌屬(Sporolactobacillus).

Anaerobic and facultative anaerobic bacteria were identified and isolated from songhe grain fermented for 30 days, strains were isolated by enrichment culturing and dilution coating plate, The best dilution and anaerobic cultured meth- od was determined by counting colony, as well, strains isolated were identified with the method of observing physical charac- terization and testing biochemical properties. The results showed that the optimal dilution used to separate anaerobic and fac- ultative anaerobic bacteria was 10-3 , the cultured method of pumping vacuum was suitable for culturing and isolating anaero bic bacteria, 5 caproic acid bacteria and 6 butyric acid bacteria which gram stain were positive were obtained and identified as Clostridium and Bacillus, 2 methane bacteria with negative gram staining were isolated and identified as Pseudomonas, 6 lactic acid bacteria which gram stain were positive were obtained and identified as Sporolactobacillus
