目的通过组织学、免疫组化方法和Calpain活性的测试,观察扇贝寡肽对运动大鼠停训致肌萎缩后肌肉湿重、肌细胞形态、肌纤维直径、横截面积、Calpain活性、肌球蛋白表达以及肌细胞凋亡过程的影响,研究扇贝寡肽对停训后骨骼肌萎缩的干预作用。方法40只SD雄性大鼠以15m/min的速度,持续性水平跑台训练30min,运动6d/week,休息1d/week,其后以速度递增3m/min/week,运动时间递增30min/week,在为期3周的训练后即随机抽取10只大鼠为基础对照组(A,n=lO),其余大鼠为B组用于制备骨骼肌萎缩模型,3周后随机分组为补充扇贝寡肽组(B,,n=10)、补充安慰剂组(B2,n=10)、补充牛乳组(B3,n=10)。实验组所有动物每天分别进行灌胃15%扇贝寡肽饮料2ml、B,组大鼠灌胃等量的牛乳,B2组大鼠灌胃等量的安慰剂。结果固定大鼠左侧后肢3周后可导致骨骼肌废用性萎缩,废用性肌萎缩的发生、发展与Calpain活性升高、水解骨骼肌收缩蛋白有关,而且,骨骼肌细胞凋亡增加在废用性肌萎缩的发生发展过程中产生重要作用。而补充扇贝寡肽可显著减低骨骼肌细胞凋亡指数,明显抑制骨骼肌Bax蛋白的表达、促进Bcl一2蛋白的表达,抑制Calpain活性,减少收缩蛋白的降解,维持肌球蛋白结构的完整,从而干预肌萎缩的进程。

목적통과조직학、면역조화방법화Calpain활성적측시,관찰선패과태대운동대서정훈치기위축후기육습중、기세포형태、기섬유직경、횡절면적、Calpain활성、기구단백표체이급기세포조망과정적영향,연구선패과태대정훈후골격기위축적간예작용。방법40지SD웅성대서이15m/min적속도,지속성수평포태훈련30min,운동6d/week,휴식1d/week,기후이속도체증3m/min/week,운동시간체증30min/week,재위기3주적훈련후즉수궤추취10지대서위기출대조조(A,n=lO),기여대서위B조용우제비골격기위축모형,3주후수궤분조위보충선패과태조(B,,n=10)、보충안위제조(B2,n=10)、보충우유조(B3,n=10)。실험조소유동물매천분별진행관위15%선패과태음료2ml、B,조대서관위등량적우유,B2조대서관위등량적안위제。결과고정대서좌측후지3주후가도치골격기폐용성위축,폐용성기위축적발생、발전여Calpain활성승고、수해골격기수축단백유관,이차,골격기세포조망증가재폐용성기위축적발생발전과정중산생중요작용。이보충선패과태가현저감저골격기세포조망지수,명현억제골격기Bax단백적표체、촉진Bcl일2단백적표체,억제Calpain활성,감소수축단백적강해,유지기구단백결구적완정,종이간예기위축적진정。
Objective Through histology and immunochemistry methods and the test of Calpain activity, this paper studies the effects of scallop peptides supplementation on muscle morphology, the Calpain activity, expression of Myosin and muscle cell apoptosis, after detraining. Methods 40 Male Sprague - Dawley rats ran 30 minutes at the speed of 15m/min , exercise 5day/ week and rest 1 day/week in the first week. In second week, rats ran 60 minutes at the speed of 18m/min, exercise 5day/week and rest lday/week and ran 90 minutes at the speed of 21m/min, exercise 5day/week and rest lday/week in the third week. After sports training for 3 weeks, ten rats were baseline group. The 30 rats were divided randomly into 3 groups as placebo, milk and SPs supplemented, then the left hind limbs of the rats were fixed by plastic gypsum for 3 weeks. Rats in groups B3 were fed 2ml beverage containing 15% SPs, and rats in control were fed 2ml placebo and milk respectively. Results The resuits showed that skeletal muscle were atrophy, after left hind limbs fixed for three weeks. Activated Calpain and hydrolyzed myosin took important roles in the process of muscular atrophy events. After SPs supplementation, the loss of mass for muscle, the Calpain activity alleviated obviously, the integrity of the myosin structure was maintained and the expression for Bcl - 2 increased obviously. But the expression for Bax decreased obviously.
