目的:研究长期低频电刺激对大鼠比目鱼肌废用性肌萎缩的影响,并初步探讨其调控机制。方法:以大鼠比目鱼肌为研究对象,采用后肢制动法建立废用性肌萎缩模型,SDH酶法检测长期低频电刺激对大鼠比目鱼肌肌纤维的影响,Real Time—PCR方法观察肌球蛋白重链Ⅰ、Ⅱa、Ⅱx基因表达变化,Western blot检测p-JNK、p—ERK1/2、p—p38激酶蛋白表达。结果:低频电刺激30日后,大鼠比目鱼肌废用性肌萎缩情况明显好转,废用性肌萎缩模型中Ⅱ型肌纤维向Ⅰ型肌纤维的转化趋势得以逆转;肌球蛋白重链Ⅰ、Ⅱa、Ⅱx基因表达变化同样显示由Ⅱ型向Ⅰ型转化的趋势;MAPK通路相关蛋白p-JNK、p-ERKl/2、p—p38均被激活。结论:长期低频电刺激可有效改善废用性肌萎缩,此过程可能与MAPK通路激活相关。

목적:연구장기저빈전자격대대서비목어기폐용성기위축적영향,병초보탐토기조공궤제。방법:이대서비목어기위연구대상,채용후지제동법건립폐용성기위축모형,SDH매법검측장기저빈전자격대대서비목어기기섬유적영향,Real Time—PCR방법관찰기구단백중련Ⅰ、Ⅱa、Ⅱx기인표체변화,Western blot검측p-JNK、p—ERK1/2、p—p38격매단백표체。결과:저빈전자격30일후,대서비목어기폐용성기위축정황명현호전,폐용성기위축모형중Ⅱ형기섬유향Ⅰ형기섬유적전화추세득이역전;기구단백중련Ⅰ、Ⅱa、Ⅱx기인표체변화동양현시유Ⅱ형향Ⅰ형전화적추세;MAPK통로상관단백p-JNK、p-ERKl/2、p—p38균피격활。결론:장기저빈전자격가유효개선폐용성기위축,차과정가능여MAPK통로격활상관。
The aim of this paper is to study the effect of low frequency electrical stimulation on the soleus muscles fibers in rats and to have a preliminary discussion of the regulatory mechanism. The method treated the rat's musculi soleus as the objects. The study adopted everyday LF electrical stimulation on the model of muscular atrophy of disuse 30 days after which it can be observed that the effect of transformation of the long term LF electrical stimulation of soleus muscular fiber under the SDH enzymic method; using the Real-time PCR method to observe the changes of myosin heavy chain (MHC) I, Ⅱa, Ⅱx genetic expression; Western blotting assay examined MAPK kinase protein expression. The results showed that after 30 days of LF electrical stimulation (ES), the muscular atrophy of disuse was clearly better as it had a good reversion of the rat's soleus muscles of model Ⅱ transforming to model I; JNK/ MAPK pathway related protein p-JNK, p-ERK1/2, p-p38 were all activated. The conclusion is that the long term LF electrical stimulation can improve the muscular atrophy of disuse effectively and the process may result from the activation of JNK/MAPK pathway.
