目的:探讨不同低氧浓度暴露对慢性疲劳综合征小鼠肠道γδT细胞的影响.方法:雄性ICR小鼠72只,随机分为6组,每组12只,即正常对照组(C0组)、慢性疲劳综合征建模组(CFS0组)、低氧干预正常对照组(C组)、慢性疲劳综合征低氧干预对照组(CFS组)、慢性疲劳综合征+15%氧浓度低氧暴露组(15%H组)和慢性疲劳综合征+18%氧浓度低氧暴露组(18%H组),其中CFS0组、CFS组、15%H组和18%H组采用束缚和强迫游泳方式建立慢性疲劳综合征小鼠模型,C0组和C组在同等条件下正常饲养.建模完成后C0组和CFS0组用于检验建模效果.C组继续饲养7d,CFS组自然恢复7d,15%H组和18%H组分别在15%和18%氧浓度环境暴露7 d(1 h/d),之后,采用流式细胞术检测各组小鼠肠道γδT细胞(CD3+ γδT+)变化.结果:1)CFS建模第1周C0组和CFS0组小鼠体重不存在显著性差异,CFS建模第2、3、4、5周,CFS0组小鼠体重明显低于C0组(P<0.05);2)水迷宫显示CFS0组潜伏期较C0组长,空间搜索实验中在平台所在象限停留时间较C0组短;3)与C组相比,CFS组和18%H组的γδT细胞在CD3+ iIEL细胞中的百分比显著下降,15%H组γδT细胞在CD3+ iIEL细胞中的百分比显著高于CFS组和18%低氧干预组.结论:小鼠体重及水迷宫相关结果提示通过束缚和强迫游泳方式成功建立慢性疲劳综合征小鼠模型;在氧浓度为15%环境中暴露7 d(1 h/d)可明显改善慢性疲劳综合征小鼠肠道γδT细胞的表达量.

Objective: It is to study the effect of different hypoxia concentration exposure on γδT cells expression in intestine in mice with chronic fatigue syndrome (CFS). Methods: 72 ICR male mice were randomly divided into 6 groups (12 per group), the model control group (Co), the chronic fatigue syndrome model group (CFS0), the hypoxia intervention control group (C), the hypoxia intervention control group with chronic fatigue syndrome (CFS), the chronic fatigue syndrome with 15% 02 concentration hypoxia exposure intervention group (15% H) and the chronic fatigue syndrome with 18% 02 concentration hypoxia exposure intervention group (18%H). Among them, the CFS0 group, CFS group, 15% H group and 18% H group were induced by restraint and forced swimming, and Co group and C group were fed in the same condition. After the establishment of CFS model, Co group and CFS0 group were used to test the effect of CFS model. C group continued to feed for 7 days, and CFS group recovered for 7 days; 15%H group and 18% H group were respectively exposed to 15% 02 concentration hypoxia and 18% 02 concentration hypoxia for 7 days ( one hour per day). The number of γδT cells in intestine was examined by the flow cytometer. Results: 1 ) There were no significant difference in the body weight between CO group and CFS0 group during the first week of the establishment of mice model, but body weight in CFS0 group was significantly lower than that in CO group in the following 2, 3, 4, 5 weeks. 2 ) Morris water maze (MWM) results showed that the incubation period of CFS0 group was shorter compared with the CO group. In addition, the platform quadrant time in CFSo group was longer compared with CO group. 3 ) The percentage of γδT cells in CD^3+ ilEL cell (intestinal intraepithelial lymphocytes) in CFS group and 18% H group decreased significantly, compared with C group, however, the percentage of T cells in CD^3+ ilEL cell in 15% H group was significantly higher than that of CFS group and 18% H group, and had no difference with C. Conclusions: Based on the change in the general body weight and behavioral indicators, the model of chronic fatigue syndrome has been successfully established by restraint and forced swimming. Moreover, the exposure to 15% O2 hypoxia for 7 days (1 hour/day) can significantly improve the expression of γδT cells in intestine in the chronic fatigue syndrome.
