目的:研究运动对RBP4诱导的胰岛素抵抗大鼠外周血中IL-2、IL-2R的表达以及CD4、CD8活性的影响.方法:8周龄雄性SD大鼠给予重组RBP4 3 μg/g体重腹腔注射,12小时注射一次,持续注射4周.设RBP4+运动组(RE)、RBP4安静组(RR)和正常安静对照组(C),RE组予4周无负重游泳训练,60 min/天.免疫组化和免疫印迹法分别检测血清RBP4,胰岛素抵抗指数(HOMA-IR).ELISA检测血清IL-2、IL-2R的表达;S-P一步法检测血液中CD4、CD8的活性.结果:RE组血清RBP4和HOMA-IR显著低于RR组.RE组较RR组大鼠CD4细胞数增多(P<0.05),CD8细胞数稍有减少(P<0.05),CD4/CD8比值明显增加(P<0.01);血清中IL-2水平明显升高(P<0.05),IL-2R水平降低(P<0.05),因而比值明显增加(P<0.01);RE组与RR组比胸腺指数(P<0.05)、脾脏指数明显提高(P<0.01).结论:RBP4诱导的胰岛素抵抗大鼠免疫功能有所下降.而运动能够明显提升RBP4诱导的胰岛素抵抗大鼠免疫能力.

목적:연구운동대RBP4유도적이도소저항대서외주혈중IL-2、IL-2R적표체이급CD4、CD8활성적영향.방법:8주령웅성SD대서급여중조RBP4 3 μg/g체중복강주사,12소시주사일차,지속주사4주.설RBP4+운동조(RE)、RBP4안정조(RR)화정상안정대조조(C),RE조여4주무부중유영훈련,60 min/천.면역조화화면역인적법분별검측혈청RBP4,이도소저항지수(HOMA-IR).ELISA검측혈청IL-2、IL-2R적표체;S-P일보법검측혈액중CD4、CD8적활성.결과:RE조혈청RBP4화HOMA-IR현저저우RR조.RE조교RR조대서CD4세포수증다(P<0.05),CD8세포수초유감소(P<0.05),CD4/CD8비치명현증가(P<0.01);혈청중IL-2수평명현승고(P<0.05),IL-2R수평강저(P<0.05),인이비치명현증가(P<0.01);RE조여RR조비흉선지수(P<0.05)、비장지수명현제고(P<0.01).결론:RBP4유도적이도소저항대서면역공능유소하강.이운동능구명현제승RBP4유도적이도소저항대서면역능력.
Objective: To study the effects of exercise on IL -2, IL -2R expression and CD4, CD8 activity in blood of RBP4 - induced insulin resistance rats. Methods:8 - week - old SD male rats were established by intraperitoneal injection with recombination RBP4 3 txg/g,2 times per day for 4 weeks. Model rats were divided into RBP4 + exercise group ( RE ) , RBP4 + rest group ( RR), and normal control group (C). RE group rata swam without load,60 rain per day for four weeks. IHC and western blotting were used to detect serum RBP4 and HOMA - IR. ELISA was used to detect serum IL-2,IL-2R expression. S -P method was used to detect activity of CIM and CD8 in the blood. Results:Serum RBP4 and HOMA -IR in RE group were significantly lower than RR group. Comparing to RR group, CIM increased(P 〈0.05) while CD8 decreased(P 〈0.05) in RE group, and CD4/CD8 ratio increased obviously(P 〈0.01 ) in RR group. IL- 2 expression grew obviously(P 〈 0. 05) while IL-2R reduced(P 〈0.05), so the IL-2/IL-2R ratio increased obviously(P 〈0.01).Thymus index(P 〈 0.05 ) and spleen index( P 〈 0.01 )in RE group rise distinctly when compare with RR group. Conclusion: Immune function of RBP4 - induced insulin resistance rats had been weakend. But exercise can strengthen the immune function of them.
