文章通过传统CTAB法、改良CTAB法、SDS法、高盐低pH法和试剂盒法等5种方法,对菊叶香藜进行基因组DNA提取,旨在筛选一种菊叶香藜基因组DNA的适宜提取方法。同时用紫外分光光度法和琼脂糖凝胶电泳对5种方法所提DNA进行检测,并对改良CTAB法提取的DNA进行RAPD-PCR扩增检测。结果表明:改良CTAB法提取速度较快,便于操作,提取的DNA质量较好,进行的RAPD-PCR扩增条带清晰,能满足分子标记的要求。因此,改良CTAB法为菊叶香藜基因组DNA可靠并合适的提取方法。

The research aimed to find out a suitable method for genomic DNA extraction from Chenopodium foetidum. The genomic DNA of Chenopodium foetidum were extracted with five genomic DNA extraction methods,namely traditional CTAB, improved CTAB, SDS, high concentration salt and reagent kit. The extracted genomicDNA samples of five methods were tested by using ultraviolet spectrophotometer and agarose gel electrophoresismethods, respectively. The DNA extracted by the improved CTAB method was detected through RAPD-PCR am-plification. The results showed that the improved CTAB method was more efficient and easy to operate. The im-proved CTAB method can extract high quality DNA and RAPD-PCR amplified DNA has clear bands. The quali-ty of extracted DNA with improved CTAB method could sufficiently meet the requirements for further molecularmarker experiment, and the improved CTAB method showed to be a reliable and suitable method for DNA extrac-tion from Chenopodium foetidum.
