以多糖提取率为评价指标,采用分光光度法测定溪黄草多糖的含量,在单因素试验的基础上,通过正交优化考察了微波功率、微波时间、料液比对多糖提取率的影响.结果表明,各因素对溪黄草多糖提取率的影响顺序由大到小依次为:微波功率,微波时间,料液比.最佳提取条件为:微波功率为中火,微波时间为25min,料液比为1:15(g/mL).在此条件下.溪黄草多糖的提取率为15.32%.与传统的索氏提取法相比,微波提取法具有提取效率高、节省时间、操作简便、易工业化等优点.

The optimal extraction technology of polysaccharides from Rabdosia serra (Maxim) Hara was studied by microwave method. The polysaccharides content was determined by visible spectrophotometry method. Based on the single factor experiment, the effects of microwave power, microwave treatment time and the ratio of solid to liquid on polysaccharides extraction efficiency were investigated further by orthogonal tests. The result showed that the effects of each factor on the extraction effects of polysaccharide in order were as follows: ex- traction power〉extraction time〉solid-liquid ratio. And the optimal conditions were microwave power of middle fire for 25 min with solid-liquid ratio 1:20 (g/mL). Under these conditions, the extraction rate of polysaccharides from Rabdosia serra (Maxim) Hara is 15.32%. Compared to the Soxhlet extraction, Microwave extraction method has some advantages such as more efficient extracting, time saving, easy to control and easy to be industrialized.
