目的:探讨8周有氧运动对APP/PS1转基因小鼠皮层AMPA受体亚型GluR1的作用,为运动促进脑功能提供实验依据。方法:2月龄清洁级APP/PS1双转基因小鼠和C57BL/6J小鼠各40只,随机分为APP/PS1安静组(AS)、APP/PS1运动组(AE)、C57安静组(CS),C57运动组(CE)。运动组小鼠进行8周的跑台运动,5 d/周,60 min/d。前10 min运动速度15 m/min,后50 min运动速度18 m/min,跑台坡度为0%。跳台回避实验检测其行为学变化,记录小鼠潜伏期、错误反应次数及达标率。免疫组化方法检测皮层突触后致密物PSD-95。Western blot检测皮层AMPA受体GluR1亚基单位的表达及GluR1-Ser831和GluR1-Ser845磷酸化位点的活性。结果:1)行为学:4月龄AS组与CE组行为学变化不明显;8周有氧运动使CE组和AE组的潜伏期延长、达标率增加(P〈0.05)。2)4月龄APP+PS1小鼠皮层PSD-95较正常对照组减少;8周有氧运动明显增加其皮层PSD-95含量。3)4月龄APP/PS1小鼠皮层AMPA GluR1受体含量及活性较正常对照组变化不大。8周运动后CE和AE组其皮层受体表达及磷酸化水平均较安静组增加。结论:4月龄的APP/PS1双转基因小鼠学习记忆整体行为学虽然无明显下降,但细胞水平皮层突触密度减少,PSD-95的组分AMPA受体GluR1活性下降。8周有氧运动提高小鼠跳台回避能力,同时明显增加皮层PSD-95含量及GluR1含量和活性。

목적:탐토8주유양운동대APP/PS1전기인소서피층AMPA수체아형GluR1적작용,위운동촉진뇌공능제공실험의거。방법:2월령청길급APP/PS1쌍전기인소서화C57BL/6J소서각40지,수궤분위APP/PS1안정조(AS)、APP/PS1운동조(AE)、C57안정조(CS),C57운동조(CE)。운동조소서진행8주적포태운동,5 d/주,60 min/d。전10 min운동속도15 m/min,후50 min운동속도18 m/min,포태파도위0%。도태회피실험검측기행위학변화,기록소서잠복기、착오반응차수급체표솔。면역조화방법검측피층돌촉후치밀물PSD-95。Western blot검측피층AMPA수체GluR1아기단위적표체급GluR1-Ser831화GluR1-Ser845린산화위점적활성。결과:1)행위학:4월령AS조여CE조행위학변화불명현;8주유양운동사CE조화AE조적잠복기연장、체표솔증가(P〈0.05)。2)4월령APP+PS1소서피층PSD-95교정상대조조감소;8주유양운동명현증가기피층PSD-95함량。3)4월령APP/PS1소서피층AMPA GluR1수체함량급활성교정상대조조변화불대。8주운동후CE화AE조기피층수체표체급린산화수평균교안정조증가。결론:4월령적APP/PS1쌍전기인소서학습기억정체행위학수연무명현하강,단세포수평피층돌촉밀도감소,PSD-95적조분AMPA수체GluR1활성하강。8주유양운동제고소서도태회피능력,동시명현증가피층PSD-95함량급GluR1함량화활성。
Purpose: This study aims to investigate the effect of 8 weeks aerobic exercise on AMPA receptor subunit GluR1 in cortex areas of APP /PS1 transgenic mice. Methods: Forty two-month old APP /PS1 transgenic mice and forty C57 BL /6J mice were randomly divided into APP /PS1 sedentary group( AS),APP /PS1 exercise group( AE),C57 sedentary group( CS) and C57 exercise group( CE). The exercise groups' mice participated in 8weeks treadmill exercise,the speed was 15 m /min during the first 10 minutes,18 m /min during the following 50 minutes,and the slope was 0%. The behavioral change was detected by step-down fear conditioning. Postysynaptic densities- 95( PSD- 95) was tested by immunohistochemistry,expression of AMPA receptor subunit GluR1,and activity of GluR1- Ser831 and GluR1- Ser845 were measured by Western blot. Results: 1) AS and CE mice's behavior doesn't change while CE and AE mice's behaviors,incubation period and correct rate increase after 8 weeks aerobic exercise( P〈0. 05). 2) PSD- 95 expression of four-month mice in AE is lower than that in CS group,but it is higher than in AE group after exercise. 3) Compared with control group,there is no change in content and activity of AMPA receptor GluR1 in exercise group. The expressions of GluR1 and the phosphorylationlevel increase significantly in AE and CE after 8 weeks aerobic exercise. Conclusions: Learning and memory abilities have no change in four months-old APP /PS1 transgenic mice,while cortical synaptic density and PSD- 95 receptor GluR1 activity decreased. Eight weeks aerobics can improve the step-down avoidance ability of mice,and increase PSD- 95 expression and GluR1 expression and activity in cortex.
