探讨CETP基因rs708272多态位点与肥胖青少年运动调节血脂效果的关联性,并通过双荧光素酶报告系统研究其分子调控机制。对象和方法:50名(男23,女27;平均年龄14.9±3.35岁)中国北方肥胖青少年进行1个月的运动减脂,以有氧运动为主辅以力量和拉伸训练,统一饮食。减脂前后取空腹静脉血测定血清TC、TG、HDL-C、LDL-C水平,PCR-RFLP法进行CETP基因rs708272分型,观察rs708272与肥胖青少年运动调节血脂效果的关联性;通过构建重组质粒,利用双荧光素酶报告系统检测重组质粒中荧光素酶报告基因的相对表达活性。结果:(1)训练后,B1B2基因型组血清TC(0.64±0.51mmol/L)、LDL-C(0.51±0.41mmol/L)的降低程度显著大于B1B1基因型组(TC:0.20±0.53mmol/L;LDL-C:0.24±0.35mmol/L);B2B2基因型组(0.10±0.15mmol/L)和B1B1基因型组(0.18±0.26mmol/L)血清HDL-C的升高程度显著大于B1B2基因型组(-0.03±0.16mmol/L)。(2)含B1等位基因的重组载体中报告基因的相对活性(11.40±0.26)显著低于B2等位基因(31.19±1.50),但两者均显著高于对照组(3.78±0.11)。结论:(1)rs708272位点多态性与运动减脂后血清TC、HDL-C、LDL-C浓度的变化程度存在一定的关联,可作为运动降血脂效果敏感性的分子遗传学标记;(2)rs708272位点处由B1→B2的突变是功能性突变,能显著提高CETP基因的表达活性。

The objective of this study was to analyze the correlation between the single nucleotide polymorphism of rs708272 loci in cholesteryl ester transfer protein(CETP) gene and changes of serum lipid in obese adolescents, and to study the molecule mechanism. Fifty obese adolescents (23 males and 27 females with an average age of 14.9±3.35) from North China took a month-long test based on aerobic exercises complemented with strength and stretch training to lose weight, and the same diet. Before and after the test, the fasting venous blood was drawn for determining plasma lipid indicators and extracting genomic DNA, then for de- tecting the luciferase report gene expression activity of recombinant plasmid. It was shown that: (1) After the aero- bic exercise, BIB2 genotype obese adolescents group's plasma TC (0. 64±0.51 mmol/L) and LDL-C (0. 51 ±0. 41 mmol/L) decreased more greater than B1B1 (TC 0. 20 ±0. 53 mmol/L;LDL-C: 0. 24± 0. 35 mmol/L). The level of plasma HDL-C of B2B2 genptype group (0. 10±0. 15 mmol/ L) and B1B1 genotype group (0. 18±0. 26 mmol/L) increased higher than B1B2 (-0. 03±0. 16mmol/L). (2) The relative activity of the luciferase reporter containing B1 allele (11.40±0. 26) of rs708272 polymorphism was significantly lower than the B2 allele (31. 19~1.50), and both were significantly higher than those of the control group (3. 78±0. 11). It could be concluded that rs708272 polymorphism correlated with exercise induced the changes of serum TC, HDL-C, LDLC in obese adolescents, and can be used as molecular genetic markers in weight loss field. The first intron of CETP gene played a positive role in the regulation of gene expression, and the mutation from B1 to B2 of rs708272 loci was a functional mutation, which could significantly increase the activity of CETP gene expression.
