目的:通过观察一次性力竭运动过程中大鼠苍白球内侧部(GPi)神经递质谷氨酸(Glu)和γ-氨基丁酸(GABA)及相关受体表达水平的动态变化,探讨力竭运动过程中苍白球内侧部神经元电活动变化的可能机制。方法:选取雄性Wistar大鼠为实验对象,采用微透析-高效液相色谱联用技术,观察一次性力竭运动过程中大鼠苍白球内侧部神经元胞外Glu和GABA水平动态变化;采用免疫荧光双标技术,检测安静、力竭时和恢复90 min时谷氨酸离子型2B受体(NR2B)和γ-氨基丁酸A型受体α1亚基(GABAAα-1)表达水平。结果:自主运动期大鼠苍白球内侧部神经元胞外Glu/GABA比值均显著下降(P〈0.05),而力竭即刻其胞外Glu/GABA比值则显著升高(P〈0.01)。与安静状态相比,苍白球内侧部在恢复90 min时NR2B受体阳性细胞数量显著增加(P〈0.05),力竭即刻和恢复90 min时GABAAα-1受体阳性细胞数量显著减少(P〈0.05,P〈0.01)。结论:力竭运动过程中大鼠苍白球内侧部Glu、GABA浓度及NR2B、GABAAα-1受体蛋白表达的改变是导致该核团神经元兴奋性变化的原因之一。
目的:通過觀察一次性力竭運動過程中大鼠蒼白毬內側部(GPi)神經遞質穀氨痠(Glu)和γ-氨基丁痠(GABA)及相關受體錶達水平的動態變化,探討力竭運動過程中蒼白毬內側部神經元電活動變化的可能機製。方法:選取雄性Wistar大鼠為實驗對象,採用微透析-高效液相色譜聯用技術,觀察一次性力竭運動過程中大鼠蒼白毬內側部神經元胞外Glu和GABA水平動態變化;採用免疫熒光雙標技術,檢測安靜、力竭時和恢複90 min時穀氨痠離子型2B受體(NR2B)和γ-氨基丁痠A型受體α1亞基(GABAAα-1)錶達水平。結果:自主運動期大鼠蒼白毬內側部神經元胞外Glu/GABA比值均顯著下降(P〈0.05),而力竭即刻其胞外Glu/GABA比值則顯著升高(P〈0.01)。與安靜狀態相比,蒼白毬內側部在恢複90 min時NR2B受體暘性細胞數量顯著增加(P〈0.05),力竭即刻和恢複90 min時GABAAα-1受體暘性細胞數量顯著減少(P〈0.05,P〈0.01)。結論:力竭運動過程中大鼠蒼白毬內側部Glu、GABA濃度及NR2B、GABAAα-1受體蛋白錶達的改變是導緻該覈糰神經元興奮性變化的原因之一。
목적:통과관찰일차성력갈운동과정중대서창백구내측부(GPi)신경체질곡안산(Glu)화γ-안기정산(GABA)급상관수체표체수평적동태변화,탐토력갈운동과정중창백구내측부신경원전활동변화적가능궤제。방법:선취웅성Wistar대서위실험대상,채용미투석-고효액상색보련용기술,관찰일차성력갈운동과정중대서창백구내측부신경원포외Glu화GABA수평동태변화;채용면역형광쌍표기술,검측안정、력갈시화회복90 min시곡안산리자형2B수체(NR2B)화γ-안기정산A형수체α1아기(GABAAα-1)표체수평。결과:자주운동기대서창백구내측부신경원포외Glu/GABA비치균현저하강(P〈0.05),이력갈즉각기포외Glu/GABA비치칙현저승고(P〈0.01)。여안정상태상비,창백구내측부재회복90 min시NR2B수체양성세포수량현저증가(P〈0.05),력갈즉각화회복90 min시GABAAα-1수체양성세포수량현저감소(P〈0.05,P〈0.01)。결론:력갈운동과정중대서창백구내측부Glu、GABA농도급NR2B、GABAAα-1수체단백표체적개변시도치해핵단신경원흥강성변화적원인지일。
Objective:The purpose of this study was to reveal the possible mechanism of the neurons activity change in globus pallidus(GPi) during exhaustive exercise by observing the dynamic changes of glutamate(Glu),γ-aminobutyric acid(GABA) and their receptors in internal segment in rats ' GPi during exhaustive exercise.Methods:Male Wistar rats were selected in this study.The expressions of the Glu and GABA in GPi during an exhaustive exercise were observed by microdialysis-high performance liquid chromatogram techniques.The expressions of NR2 B receptor and GABAAα-1 receptor were detected at rest,during exercise,90 min after exercise,by double-lable immunofluorescence technique.Results:Compared with rest state,the Glu / GABA ratio of GPi significantly decreased during the auto-exercise period(P 0.05),while it was significantly increased at exhaustion time point(P 0.01).The number of NR2 B positive cells in GPi was increased significantly at 90 min after exhaustive exercise compared with rest state(P 0.05),but number of GABAAα-1 positive cells was decreased significantly both at exhaustion time point(P 0.05) and 90 min after exhaustion(P 0.01).Conclusion:The change of expressions of Glu,GABA,NR2 B receptor and GABAAα-1 receptor in rats' GPi during exhaustive exercise is a reason for neurons activity change.
