目的 研究CD4+IL-9+T细胞因子IL-9、IL-10在大肠癌肿瘤免疫微环境中的表达水平,探讨其与临床病理的相关性. 方法 采集2014年6月1日至2014年10月8日,本院初次行手术治疗的45例大肠癌患者的肿瘤组织、对应癌旁组织及外周血(术前和术后),同时取15例健康志愿者外周血液.运用液相蛋白芯片检测技术检测上述组织和外周血中CD4+IL-9+T细胞主要细胞因子IL-9、IL-10的浓度.结果 细胞因子IL-9在大肠癌患者微环境中的浓度为(0.82±0.51)pg/ml,高于癌旁(0.56±0.52)pg/ml,P=0.0318;同时,它也高于术前、术后及健康人外周血中IL-9的浓度,P<0.05;细胞因子IL-10在大肠癌患者微环境中的浓度高于癌旁,分别为(0.87±0.43),(0.52±0.49)pg/ml,P<0.05,也高于术前、术后及健康人外周血中IL-10的浓度,P<0.05. 结论 大肠癌肿瘤微环境中的CD4+IL-9+T细胞高表达细胞因子IL-9、IL-10,且细胞因子IL-9表达水平与肿瘤Duke' s分期存在相关性,分期越高IL-9的蛋白水平越高.由此可见干扰IL-9、IL-10的信号通路或活性或许能够成为治疗大肠肿瘤的新靶点.

Objective To investigate the expression of CD4+IL-9+T cytokines IL-9 and IL-10 in colorectal tumor immune micro-environment, and the relationship with clinical pathology. Methods Collected 45 cases of colorectal cancer tumor tissues and corresponding para cancer tissues and peripheral blood(before and after surgery, initial surgery treatement in our hospital between June 1 and October 8, 2014),and 15 healthy volunteers in pe-ripheral blood. The concentration of main cytokines IL-9 and IL-10 of CD4+IL-9+T cells in the tissue and peripheral blood were detected using liquid pro-tein chip detection technology. Results The concentration of IL-9 in the tumor micro-environment (0.82±0.51)pg/ml was higher than that in the adja-cent tissue (0.56±0.52)pg/ml, P=0.0318, it was also higher than that in the before or after operation,and healthy human peripheral blood, P<0.05. IL-10 in the tumor micro-environment was higher than that in the adjacent tissue, P<0.05,was also higher than that in the before or after operation and healthy human peripheral blood,P<0.05. Conclusion The cytokines IL-9, IL-10 of CD4+IL-9+T cells was highly expressed in the tumor micro-environment, correla-tion test showed that protein level of IL-9 was correlated with Duke's stage, higher stages with higher IL-9 level. Interference signal pathway or activity of IL-9, IL-10 may become a new target in the treatment of rectal cancer.
