目的 探讨唑来膦酸干预原发性骨质疏松( OP)模型鼠后骨髓脂肪含量变化规律. 方法 27只3月龄雌性SD大鼠随机均分为假手术组( SHAM)、去卵巢组( OVX)及OVX+唑来膦酸干预组( ZOL). 分别在术后0、6、12周行MRI水-脂分离成像及BMD扫描,获取骨髓脂肪分数( MFF)及BMD. 实验结束行骨髓脂肪细胞定量分析. 结果 OVX组MFF呈时序性增高而BMD呈时序性降低. 第6、12周时,OVX组MFF较基线时分别增高40.0%、69.4%( P<0.001 ). 第6周始组间MFF有显著性差异,而组间BMD至第12周时才有统计学差异(P<0.001). OVX组骨髓脂肪细胞密度、直径及面积百分比较SHAM组分别增高57.3%、29.5%、163%,较ZOL分别增高65%、22.2%、144%(P<0.001). ZOL组MFF及脂肪细胞定量参数与SHAM组无明显差异. 结论 ZOL保护骨受损同时,可能通过抑制骨髓脂肪含量而起作用.

Objective To explore the effect of zoledronic acid ( ZA) in restoring marrow adipogenesis in estrogen-deficient rats. Methods Twenty-seven female SD rats were randomly divided equally into SHAM, ovariectomized ( OVX) , and OVX+ZOL groups.Marrow fat fraction (MFF) and bone mineral density (BMD) were measured at 0, 6, and 12 weeks.Marrow adipocytes were quantitatively evaluated by histopathology.Results OVX rats exhibited time-depend increase in MFF but decrease in BMD. The increase of MFF above baseline was 40.0% at week 6 and 69.4% at week 12, respectively (P<0.001).Compared with SHAM rats, MFF in OVX rats markedly elevated at week 6 but a reduction in BMD was first detected at week 12 ( P<0.001) .The adipocyte density, diameter, and volume in OVX rats increased by 57.3%, 29.5%, and 163%, respectively, compared with those in SHAM rats, and by 65%, 22.2%, and 144%, respectively, compared with those in ZOL rats (P<0.001).MFF and adipocyte parameters were not different between ZOL rats and SHAM rats.Conclusion A single dose of ZOL is able to reverse the marrow adiposity of OVX-induced osteoporosis.
