目的 探讨绝经后骨质疏松症肾阳虚证与LTBP1蛋白表达及cDNA变化的关联性. 方法 随机选择绝经后骨质疏松症患者,中医辨证肾阳虚证组27例,27例健康绝经后妇女为对照组,Western-blot法检测外周血LTBP1的蛋白表达,对cDNA片段进行测序. 结果 肾阳虚证组LTBP1蛋白表达水平(0.365 ±0.278)低于健康对照组(0.675 ±0.583)(P<0.05),差异有统计学意义. cDNA测序在健康对照组与肾阳虚证组中碱基无有意义的突变. 结论 绝经后骨质疏松症肾阳虚证与LTBP1蛋白表达下调相关联,测序发现与碱基突变无关.

Objective To study the association between LTBP1 protein and cDNA expression and the kidney Yang deficiency syndrome in postmenopausal osteoporosis.Methods Patients with postmenopausal osteoporosis were randomly selected.Twenty-seven cases were in TCM syndrome of kidney Yang deficiency syndrome group, and 27 healthy postmenopausal women were in the control group.Protein expression of LTBP1 was detected using Western blotting method.The cDNA fragment was sequenced. Results LTBP1 protein expression level in kidney Yang deficiency syndrome group (0.365 ±0.278) was less than that in healthy controls (0.675 ±0.583), and the difference was statistically significant (P<0.05).There was no significant mutation in CDNA bases in both healthy controls and kidney Yang deficiency syndrome group.Conclusion The down-regulation of LTBP1 protein expression is associated with postmenopausal osteoporosis with kidney Yang deficiency syndrome.There is no correlation with mutation of DNA bases.
