目的 考察鼠李糖乳杆菌(LGG)对阪崎克罗诺杆菌致脑膜炎的抑制作用.方法 以Caco-2细胞为体外模型,采用黏附侵袭实验研究阪崎克罗诺杆菌对Caco-2细胞的黏附侵袭作用及最佳感染时间,并用不同浓度的LGG与Caco-2细胞作用,确定其抑制阪崎克罗诺杆菌的最佳浓度.进一步通过竞争、排斥和置换实验检测LGG对阪崎克罗诺杆菌黏附侵袭的抑制作用.通过体内实验,研究LGG对乳鼠脑膜炎的作用效果.结果 阪崎克罗诺杆菌对Caco-2细胞具有黏附侵袭性,最佳感染时间为3 h.随着加入LGG浓度的增加,阪崎克罗诺杆菌对Caco-2细胞的黏附和侵袭作用被显著抑制.LGG能通过竞争、排斥作用抑制阪崎克罗诺杆菌对Caco-2细胞的黏附.LGG对阪崎克罗诺杆菌引起的脑膜炎具有预防和治疗作用,且预防效果更显著.结论 通过体内外实验,LGG可以抑制阪崎克罗诺杆菌进入肠屏障,从而减少其穿越血脑屏障的量,最终达到预防和治疗脑膜炎的效果.
目的 攷察鼠李糖乳桿菌(LGG)對阪崎剋囉諾桿菌緻腦膜炎的抑製作用.方法 以Caco-2細胞為體外模型,採用黏附侵襲實驗研究阪崎剋囉諾桿菌對Caco-2細胞的黏附侵襲作用及最佳感染時間,併用不同濃度的LGG與Caco-2細胞作用,確定其抑製阪崎剋囉諾桿菌的最佳濃度.進一步通過競爭、排斥和置換實驗檢測LGG對阪崎剋囉諾桿菌黏附侵襲的抑製作用.通過體內實驗,研究LGG對乳鼠腦膜炎的作用效果.結果 阪崎剋囉諾桿菌對Caco-2細胞具有黏附侵襲性,最佳感染時間為3 h.隨著加入LGG濃度的增加,阪崎剋囉諾桿菌對Caco-2細胞的黏附和侵襲作用被顯著抑製.LGG能通過競爭、排斥作用抑製阪崎剋囉諾桿菌對Caco-2細胞的黏附.LGG對阪崎剋囉諾桿菌引起的腦膜炎具有預防和治療作用,且預防效果更顯著.結論 通過體內外實驗,LGG可以抑製阪崎剋囉諾桿菌進入腸屏障,從而減少其穿越血腦屏障的量,最終達到預防和治療腦膜炎的效果.
목적 고찰서리당유간균(LGG)대판기극라낙간균치뇌막염적억제작용.방법 이Caco-2세포위체외모형,채용점부침습실험연구판기극라낙간균대Caco-2세포적점부침습작용급최가감염시간,병용불동농도적LGG여Caco-2세포작용,학정기억제판기극라낙간균적최가농도.진일보통과경쟁、배척화치환실험검측LGG대판기극라낙간균점부침습적억제작용.통과체내실험,연구LGG대유서뇌막염적작용효과.결과 판기극라낙간균대Caco-2세포구유점부침습성,최가감염시간위3 h.수착가입LGG농도적증가,판기극라낙간균대Caco-2세포적점부화침습작용피현저억제.LGG능통과경쟁、배척작용억제판기극라낙간균대Caco-2세포적점부.LGG대판기극라낙간균인기적뇌막염구유예방화치료작용,차예방효과경현저.결론 통과체내외실험,LGG가이억제판기극라낙간균진입장병장,종이감소기천월혈뇌병장적량,최종체도예방화치료뇌막염적효과.
Objective To investigate the inhibitory effect of Lactobacillus rhamnosus GG ( LGG) against Cronobacter-induced meningitis in neonatal rats. Methods The cell adhesion and invasion capacities of Cronobacter were assayed in Caco-2 cells, and the optimal time length and concentration of the bacterium for infection were determined. The suppressive effects of LGG on the adhesion and invasion of Cronobacter in caco-2 cells were tested by competitive and exclusion experiments, and its inhibitory effect against Cronobacter-induced meningitis was evaluated in neonatal rats. Results Cronobacter showed aggressive adhesion to caco-2 cells with an optimal infection time of 3 h. LGG produced a concentration-dependent inhibition of Cronobacter adhesion and invasion by competing with and excluding the latter for cell adhesion. In neonatal rats, LGG showed an obvious preventive effect and also a moderate therapeutic effect against Cronobacter-induced meningitis. Conclusion LGG can inhibit Cronobacter entry across the intestinal barrier to achieve preventive and therapeutic effects against Cronobacter-induced meningitis.
