利用反相高效液相色谱法测定八角中两种黄酮的含量。采用Zorbax Eclipse C18色谱柱(4.6 mm ×150 mm,5μm ),以甲醇-0.4%磷酸(V/V,50∶50)混合溶液为流动相,在360 nm 波长处进行测定。结果表明:质量浓度在0.025~0.5 mg/mL时,槲皮素和山柰酚峰面积和浓度呈线性关系。槲皮素、山柰酚的平均回收率分别为97.45%,102.51%,RSD分别为2.3%,2.8%。在此条件下测得水解样中槲皮素0.508 mg/g (n=3,RSD=2.860%)、山柰酚0.325 mg/g (n=3,RSD=1.10%),总黄酮提取物中槲皮素0.110 mg/g (n=3,RSD=1.29%)、山柰酚0.031 mg/g (n=3,RSD=2.24%)。

The RP-HPLC is applied to determine two flavonoids,such as quercetin and kaempferol in Illicium verum.The sample solution is separated by using Zorbax Eclipse C 18 (4.6 mm×150 mm, 5μm)as stationary phase and a mixture of methanol and 0.4% phosphoric acid solution (50∶50)as mobile phase.The content of the two flavonoids is determined at the wave length of 360 nm.The peak areas and concentration of quercetin and kaempferol have a good linear relationship as the mass concentration of 0.025~0.5 mg/mL.The tests for recovery and precision are made by standard addition method.The recoveries of quercetin and kaempferol are 97.45%,102.51%and the RSDs are 2 .3%,2 .8% respectively.The content of quercetin and kaempferol in flavonoid hydrolysates is 0.508 mg/g (n= 3,RSD= 2.860%)and 0.325 mg/g (n= 3,RSD= 1.10%),0.110 mg/g (n= 3, RSD= 1.29%)and 0.031 mg/g (n= 3,RSD= 2.24%)in flavonoid extracts.
