目的:研究靛玉红、色胺酮对脂多糖(LPS)诱导的 RAW264.7炎症细胞的抗炎作用。方法以10μg/ ml LPS 刺激 RAW264.7细胞24 h 诱导 UC 体外炎症模型,以 MTT 法检测靛玉红、色胺酮的对正常细胞的作用,以酶联免疫吸附法(ELISA)研究靛玉红、色胺酮在直接干预、预防干预2种模式下对细胞培养液白细胞介素-6(IL -6)/肿瘤坏死因子(TNF -α)浓度的影响。结果模型组中 IL -6/ TNF -α浓度较正常组均有明显升高(P ﹤0.01);靛玉红直接干预时,细胞培养液 IL -6(5.0、10.0μmol/ L)/ TNF -α(2.5、5.0、10.0μmol/ L)均较模型组有明显下降(P ﹤0.05);色胺酮直接干预时,细胞培养液 IL -6(0.8、8.0μmol/ L)/ TNF -α(0.8μmol/ L)均较模型组有明显下降( P ﹤0.01,P ﹤0.05),预防作用模式下,细胞培养液 IL -6(0.08、0.8μmol/ L)较模型组有明显下降(P ﹤0.05)。结论靛玉红、色胺酮有一定的体外抗炎作用,抗炎模式以直接干预为主,抗炎机制可能为下调 IL -6/ TNF -α的表达,可用于防治溃疡性结肠炎。

목적:연구전옥홍、색알동대지다당(LPS)유도적 RAW264.7염증세포적항염작용。방법이10μg/ ml LPS 자격 RAW264.7세포24 h 유도 UC 체외염증모형,이 MTT 법검측전옥홍、색알동적대정상세포적작용,이매련면역흡부법(ELISA)연구전옥홍、색알동재직접간예、예방간예2충모식하대세포배양액백세포개소-6(IL -6)/종류배사인자(TNF -α)농도적영향。결과모형조중 IL -6/ TNF -α농도교정상조균유명현승고(P ﹤0.01);전옥홍직접간예시,세포배양액 IL -6(5.0、10.0μmol/ L)/ TNF -α(2.5、5.0、10.0μmol/ L)균교모형조유명현하강(P ﹤0.05);색알동직접간예시,세포배양액 IL -6(0.8、8.0μmol/ L)/ TNF -α(0.8μmol/ L)균교모형조유명현하강( P ﹤0.01,P ﹤0.05),예방작용모식하,세포배양액 IL -6(0.08、0.8μmol/ L)교모형조유명현하강(P ﹤0.05)。결론전옥홍、색알동유일정적체외항염작용,항염모식이직접간예위주,항염궤제가능위하조 IL -6/ TNF -α적표체,가용우방치궤양성결장염。
Objective To investigate anti - inflammatory effect of indirubin and tryptanthrin on LPS Induced RAW264. 7 inflammatory cell model. Methods Murine macrophageRAW264. 7 cells were stimula-ted by 10 μg/ ml LPS for 24 h to establish an UC inflammatory cell model. Cell viability was detected by MTT assay to explore the safe range of concentrations. UC inflammatory cell model was treated by indirubin and tryptanthrin in two different ways:treatment mode and prevention mode. IL - 6 and TNF - αin supernatant was tested by Elisa assay. Results IL - 6 and TNF - α in supernatant in model group were significantly higher than control group(P ﹤ 0. 01);Under treatment mode,IL - 6(5. 0、10. 0 μmol/ L)and TNF - α(2. 5、5. 0、10. 0 μmol/ L)in supernatant were reduced by indirubin significantly( P ﹤ 0. 05);IL - 6(0. 8、8. 0μmol/ L)and TNF - α(0. 8 μmol/ L group)in supernatant were reduced by tryptanthrin significantly(P ﹤0. 01,P ﹤ 0. 05,P ﹤ 0. 01);While under prevention mode,only IL - 6(0. 08、0. 8 μmol/ L groups)in super-natant were reduced by tryptanthrin(P ﹤ 0. 05). Conclusion Indirubin and tryptanthrin could inhibit in-flammation in UC inflammatory cell model in vitro,mainly under treatment mode by down - regulating IL - 6 and TNF - αexpression.
