检测并分析杰出有氧耐力表型与运动应激适应的表观遗传学基因调控模式特征的关联。方法:递增负荷法检测33名现役国家级耐力项目运动员和58名体育教育专业大学生(二级运动员)的最大摄氧量、个体乳酸阈,同步采用microarray及qRT‐PCR法检测训练诱导的c‐miRNA表达谱,分析杰出耐力表型与训练诱导的miRNA及去乙酰化酶(SIRT1)的基因表达调控特征的关联。结果:杰出耐力表型差异表达27条c‐miRNA ,18条上调,9条下调,其靶基因富集于AMPK、PPAR、MAPK及SIRT1信号通路,miRNA的调控作用使AM PK通路及下游通路活性上调,SIRT1的表达和活性水平显著上调并增强AM PK通路活性。结论:运动应激诱导的miRNA‐SIRT1的调控特征与杰出有氧耐力表型存在关联,对杰出耐力表型的形成具有促进作用,可作为预测有氧耐力发展潜力的表观遗传学分子标记应用于运动选材。
檢測併分析傑齣有氧耐力錶型與運動應激適應的錶觀遺傳學基因調控模式特徵的關聯。方法:遞增負荷法檢測33名現役國傢級耐力項目運動員和58名體育教育專業大學生(二級運動員)的最大攝氧量、箇體乳痠閾,同步採用microarray及qRT‐PCR法檢測訓練誘導的c‐miRNA錶達譜,分析傑齣耐力錶型與訓練誘導的miRNA及去乙酰化酶(SIRT1)的基因錶達調控特徵的關聯。結果:傑齣耐力錶型差異錶達27條c‐miRNA ,18條上調,9條下調,其靶基因富集于AMPK、PPAR、MAPK及SIRT1信號通路,miRNA的調控作用使AM PK通路及下遊通路活性上調,SIRT1的錶達和活性水平顯著上調併增彊AM PK通路活性。結論:運動應激誘導的miRNA‐SIRT1的調控特徵與傑齣有氧耐力錶型存在關聯,對傑齣耐力錶型的形成具有促進作用,可作為預測有氧耐力髮展潛力的錶觀遺傳學分子標記應用于運動選材。

The objective of the study was to explore the relationship between the elite endurance phenotype and epigenetics gene regulating effect of miRNA‐SIRT1 in exercise stress adaption .33 active elite athletes (national grade) and 58 student athletes (grade 2) were engaged in this test .VO2max and ILT were measured .Differential expression profiles of c‐miR‐NAs induced by exercise were detected .Targetscan ,miRanda ,and miRBase were used to predict target genes of those miRNA ,and pathway analysis was performed for target gene by KEGG and IPA .Result :There were 27 c‐miRNAs which regulated the pathway of AMPK ,PPAR ,MAPK ,SIRT1 ,and their regulating characters were validated to promote the formation of the elite endurance .It could be concluded that there was relationship between miRNA‐SIRT1 and exercise stress adaption ,the profile of c‐miRNAs induced by exercise could be used as an epigenetic biomarker on forecasting and valuing the potential of aerobic endurance .
