目的:研究左炔诺孕酮纳米载药缓释聚乳酸(PLA)-聚苯乙烯(PS)复合材料对体外胚胎着床的影响。方法分别观察空白对照组,空载膜组和载药膜组 Bewo 细胞球状体在 Ishikawa 单层细胞上的黏附率和铺展直径,检测各组 Ishikawa 细胞整合素αv mRNA 和蛋白的表达。结果与空载膜组和空白对照组相比,载药膜组 Bewo 细胞球状体的黏附率和铺展直径降低,Ishikawa 细胞整合素αv 的 mRNA 和蛋白表达水平均下降,差异均有统计学意义。与空白对照组相比,空载膜和载药膜均不影响 Ishikawa 细胞的增殖。结论空载膜不影响体外胚胎的着床,载药膜抑制了体外胚胎的着床。

목적:연구좌결낙잉동납미재약완석취유산(PLA)-취분을희(PS)복합재료대체외배태착상적영향。방법분별관찰공백대조조,공재막조화재약막조 Bewo 세포구상체재 Ishikawa 단층세포상적점부솔화포전직경,검측각조 Ishikawa 세포정합소αv mRNA 화단백적표체。결과여공재막조화공백대조조상비,재약막조 Bewo 세포구상체적점부솔화포전직경강저,Ishikawa 세포정합소αv 적 mRNA 화단백표체수평균하강,차이균유통계학의의。여공백대조조상비,공재막화재약막균불영향 Ishikawa 세포적증식。결론공재막불영향체외배태적착상,재약막억제료체외배태적착상。
Objective To examine the effect of levonorgestrel-loaded nanoparticle composites on embryo implantation in vitro. Methods We detected the Bewo cell spheroids’adhesion rate and spreading rate as well as the mRNA and protein of integrin αv in the control group,the empty biofilm group and drug-loaded biofilm group. Results Compared with control group and the empty biofilm group,the drug-loaded biofilm group showed obvious difference in Bewo cell spheroids’adhesion rate and spreading rate as well as the expression level of mRNA and protein of integrin αv of Ishikawa cells.Compared with the control group,the empty biofilm group and the drug-loaded biofilm group had no effect on proliferation of Ishikawa cells. Conclusion Polylactic acid-polystyrene composites have no effect on embryo implantation, while levonorgestrel-loaded nanoparticle polylactic acid-polystyrene composites affect embryo implantation in vitro.The data provide a theoretical basis for biological safety of empty biofilm and effectiveness of drug-loaded biofilm,and for levonorgestrel-loaded nanoparticle polylactic acid-polystyrene composites in clinical application as a new intrauterine contraceptive system.
