为了观察GnRH激动剂曲普瑞林对人乳腺癌细胞生长的影响,探讨药物处理后细胞中Fas和FasL基因的表达情况及其与细胞生长的关系,本研究采用了形态学显微镜观察、MTT检测和Real-time PCR技术,对曲普瑞林处理后乳腺癌细胞的生长增殖情况及Fas和FasL基因表达的变化进行了研究。结果显示,曲普瑞林可抑制乳腺癌细胞MCF-7的生长并表现出了剂量依赖性,药物浓度越大对肿瘤细胞的抑制作用越强。随着药物浓度的增大、抑制作用增强,肿瘤细胞中Fas基因的表达量逐渐增大,而FasL基因的表达量却逐渐下降。推测曲普瑞林在作用过程中可能通过促进肿瘤细胞中Fas的表达,诱导了Fas所介导的细胞凋亡途径的恢复,达到抑制肿瘤细胞生长的目的。
為瞭觀察GnRH激動劑麯普瑞林對人乳腺癌細胞生長的影響,探討藥物處理後細胞中Fas和FasL基因的錶達情況及其與細胞生長的關繫,本研究採用瞭形態學顯微鏡觀察、MTT檢測和Real-time PCR技術,對麯普瑞林處理後乳腺癌細胞的生長增殖情況及Fas和FasL基因錶達的變化進行瞭研究。結果顯示,麯普瑞林可抑製乳腺癌細胞MCF-7的生長併錶現齣瞭劑量依賴性,藥物濃度越大對腫瘤細胞的抑製作用越彊。隨著藥物濃度的增大、抑製作用增彊,腫瘤細胞中Fas基因的錶達量逐漸增大,而FasL基因的錶達量卻逐漸下降。推測麯普瑞林在作用過程中可能通過促進腫瘤細胞中Fas的錶達,誘導瞭Fas所介導的細胞凋亡途徑的恢複,達到抑製腫瘤細胞生長的目的。

In order to observe the effect of GnRH aganist triptorelin on the growth of human breast cancer cells, and to explore the expression of Fas and FasL gene in drug treated cells and its relationship with cell growth, the morphological microscope, MTT detection and Real-time PCR were used to study the proliferation and the expression of Fas and FasL gene of breast cancer cells which were treated by the triptorelin. The results showed that triptorelin could inhibit the growth of breast cancer cells MCF-7 and exhibited a characteristic of dose-dependent, the higher the concentration of drugs, the stronger inhibition of tumor cells. It was also observed that the higher the concentration of reagents, the ex-pression of Fas gene in breast cancer cells gradually increased, but the expression of FasL gene was gradually decreased. Therefore, it suggested that the triptorelin might promote the expression of Fas gene in tumor cells, which induced the recovery of the Fas mediated apoptosis pathway, then the growth of tumor cells were inhibited.
