目的:探讨大蒜素对人宫颈癌HeLa细胞周期和凋亡的影响。方法将2.5、5、10、20、40、80、120μg/mL大蒜素分别作用于体外培养的HeLa细胞,24、48、72 h后,于倒置相差显微镜下观察HeLa细胞形态学变化;采用WST-8法测定HeLa细胞的增殖抑制率;流式细胞术测定HeLa细胞的凋亡率和细胞周期。结果倒置相差显微镜下观察到细胞凋亡的形态学改变。大蒜素干预后HeLa细胞增殖受抑,且呈浓度与时间依赖性(P均<0.05);随着大蒜素浓度的增加,细胞凋亡率增加,G1期细胞比例增加。结论大蒜素能阻滞HeLa细胞的周期进展,诱导细胞凋亡。

목적:탐토대산소대인궁경암HeLa세포주기화조망적영향。방법장2.5、5、10、20、40、80、120μg/mL대산소분별작용우체외배양적HeLa세포,24、48、72 h후,우도치상차현미경하관찰HeLa세포형태학변화;채용WST-8법측정HeLa세포적증식억제솔;류식세포술측정HeLa세포적조망솔화세포주기。결과도치상차현미경하관찰도세포조망적형태학개변。대산소간예후HeLa세포증식수억,차정농도여시간의뢰성(P균<0.05);수착대산소농도적증가,세포조망솔증가,G1기세포비례증가。결론대산소능조체HeLa세포적주기진전,유도세포조망。
Objective To investigate the influence of allicin on the cell cycle progression and apoptosis of human cer-vical carcinoma HeLa cells.Methods The HeLa cells cultured in vitro were treated with different concentrations of allicin (2.5, 5, 10, 20, 40, 80 and 120μg/mL) for 24 h, 48 h and 72 h.Then, the morphologic changes were observed by in-verted phase contrast microscope.WST-8 was used to measure the proliferative inhibition rate of allicin on HeLa cells, and the cell cycle progression and apoptosis rate were detected by flow cytometry.Results The morphological changes of ap-optosis was observed by inverted phase contrast microscope.The proliferation of HeLa cells was inhibited after the interven-tion of Allicin, which was in a dose-and time-dependent manner(all P<0.05).With the increasing concentration of alli-cin, the apoptosis rate was increased and the proportion of G0/G1 cells was increased significantly.Conclusion Allicin can inhibit the HeLa cell proliferation, which may be related to the inducing cell apoptosis and blocking cell cycle progres-sion.
