目的：探讨隐丹参酮（CPT）对人胃癌细胞 SGC-7901凋亡的影响及其机制。方法用不同浓度的 CPT 作用于人胃癌细胞 SGC-7901，采用四甲基偶氮唑蓝（MTT）法检测细胞增殖的活力；流式细胞术检测细胞凋亡的变化；Real Time RT-PCR 法检测细胞凋亡相关蛋白 Bax、Bcl-2、p53和 p21 mRNA 表达的变化；Western blot 法检测细胞凋亡相关蛋白 Bax、Bcl-2、p53和 p21表达的变化。结果CPT 作用于人胃癌细胞 SGC-790124 h 后，可明显抑制细胞生长，诱导细胞发生凋亡；在转录水平上调促细胞凋亡蛋白 Bax 和 p53 mRNA 的表达，下调抑制细胞凋亡蛋白 Bcl-2 mRNA 的表达；在翻译水平上调促细胞凋亡蛋白 Bax 和 p53的表达，下调抑制细胞凋亡蛋白 Bcl-2的表达。结论CPT 对人胃癌细胞SGC-7901具有诱导凋亡的作用，其机制可能与线粒体途径相关。
목적：탐토은단삼동（CPT）대인위암세포 SGC-7901조망적영향급기궤제。방법용불동농도적 CPT 작용우인위암세포 SGC-7901，채용사갑기우담서람（MTT）법검측세포증식적활력；류식세포술검측세포조망적변화；Real Time RT-PCR 법검측세포조망상관단백 Bax、Bcl-2、p53화 p21 mRNA 표체적변화；Western blot 법검측세포조망상관단백 Bax、Bcl-2、p53화 p21표체적변화。결과CPT 작용우인위암세포 SGC-790124 h 후，가명현억제세포생장，유도세포발생조망；재전록수평상조촉세포조망단백 Bax 화 p53 mRNA 적표체，하조억제세포조망단백 Bcl-2 mRNA 적표체；재번역수평상조촉세포조망단백 Bax 화 p53적표체，하조억제세포조망단백 Bcl-2적표체。결론CPT 대인위암세포SGC-7901구유유도조망적작용，기궤제가능여선립체도경상관。
Objective To investigate the apoptosis effect of cryptotanshinone (CPT)on human gastric cancer cell line SGC-7901,so as to explore the molecular mechanism related.Methods SGC-7901 cells were treated with varying concentration of CPT.MTT assays and flow cytometry were used to evalu-ate the effects of CPT on the proliferation and apoptosis of SGC-7901 cells.The protein expressions of Bax,Bcl-2,p53 and p21,a series of apoptosis-related proteins,were observed by Western blotting,and the mRNA expressions of Bax,Bcl-2,p53 and p21were determined by real-time RT-PCR.Results Fol-lowing 24 h treatment,CPT was capable of inhibiting proliferation and inducing apoptosis of SGC-7901 cells significantly.In addition,Bax and p53 mRNA and protein were upregulated,while Bcl-2 mRNA and protein were downregulated.It is well known that both Bcl2 and Bax are transcriptional targets for the tumour suppressor protein,p53,which induces cell cycle arrest or apoptosis.Conclusion Apoptosis can be induced in SGC-7901 cells by CPT,potentially via a mitochondrial pathway.
