目的:比较山药芡实药对不同剂量对脾虚模型小鼠体重及免疫指标胸腺指数、T 细胞增殖能力、白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)的影响,探究山药芡实药对在提高免疫力方面的最佳剂量与配比。方法将小鼠随机分为山药芡实3∶1组、山药芡实2∶1组、山药芡实1∶1组、山药芡实1∶2组4个实验组和模型组、正常组共6组,以番泻叶水煎剂灌胃对实验组和模型组进行脾虚模型的建造,实验组以不同剂量的山药芡实药对汤剂灌胃,模型组和正常组给予等量生理盐水灌胃,称重计量体重和胸腺指数,MTT 比色法检测 T 细胞增殖能力,双抗夹心酶联免疫吸附剂测定(ELISA)法检测血清 IL-2、IL-4和 IFN-γ水平。结果造模后小鼠体重明显低于正常组(P <0.05),造模成功。给药后各实验组相关免疫指标均有不同程度改善,其中山药芡实3∶1组和山药芡实2∶1组改善程度最大,尤其是血清 IL-2、IL-4水平和胸腺指数(P <0.05,P <0.01),而在体重、T 细胞增殖能力方面,山药芡实2∶1组改善程度大于山药芡实3∶1组。结论山药芡实药对能通过影响免疫功能来改善小鼠脾虚状态,具有健脾益气的功效;推测山药芡实药对的临床最佳剂量与配比为2∶1(30 g∶15 g)。

목적:비교산약검실약대불동제량대비허모형소서체중급면역지표흉선지수、T 세포증식능력、백세포개소-2(IL-2)、백세포개소-4(IL-4)、간우소-γ(IFN-γ)적영향,탐구산약검실약대재제고면역력방면적최가제량여배비。방법장소서수궤분위산약검실3∶1조、산약검실2∶1조、산약검실1∶1조、산약검실1∶2조4개실험조화모형조、정상조공6조,이번사협수전제관위대실험조화모형조진행비허모형적건조,실험조이불동제량적산약검실약대탕제관위,모형조화정상조급여등량생리염수관위,칭중계량체중화흉선지수,MTT 비색법검측 T 세포증식능력,쌍항협심매련면역흡부제측정(ELISA)법검측혈청 IL-2、IL-4화 IFN-γ수평。결과조모후소서체중명현저우정상조(P <0.05),조모성공。급약후각실험조상관면역지표균유불동정도개선,기중산약검실3∶1조화산약검실2∶1조개선정도최대,우기시혈청 IL-2、IL-4수평화흉선지수(P <0.05,P <0.01),이재체중、T 세포증식능력방면,산약검실2∶1조개선정도대우산약검실3∶1조。결론산약검실약대능통과영향면역공능래개선소서비허상태,구유건비익기적공효;추측산약검실약대적림상최가제량여배비위2∶1(30 g∶15 g)。
Objective To compare the effects of herbal pair,Shanyao (Rhizoma Dioscoreae)and Qian-shi (Semen Euryales),at different dosage ratios on spleen-deficiency mice’s weight and immunity pa-rameters including thymus weight,proliferation of T cells as well as IL-2,IL-4 and IFN-γ,and to study the optimal dose and best ratio of this pair in terms of immunity improvement.Methods ICR mice were randomly divided into 4 experimental groups (ratios of 3 ∶1,2∶1,1 ∶1 and 1 ∶2 respectively),model group and normal group.To establish spleen-deficiency model,the experimental groups and the model group were given senna decoction by gavage,and the normal group was given equal volume of saline by gavage.Experimental groups were then given different dosages of Qianshi and Shanyao decoction by ga-vage,while the model group and the normal group were given equal volume of saline by gavage.Body Mass Index and thymus index were measured by weighing,T cell proliferation ability was tested by using MTT,and the serum levels of IL-2,IL-4 and IFN-γwere tested with a double-antibody sandwich ELISA method.Results After the establishment of spleen-deficiency model,mice’s weights were significantly below normal range (P <0.05),which demonstrated success of the model establishment.After the gava-ges of the herbal pair,associated immune indexes in all experimental groups improved at various degrees, with the most significant improvement in 3∶1 group and 2∶1 group (P <0.05),especially their serum levels of IL-2 and IL-4 as well as the thymus index.A greater degree of improvement was observed in 2∶1 group than 3∶1 group in terms of weight and T cell proliferation ability.Conclusion The herbal pair of Qianshi and Shanyao could relieve the mice’s spleen deficiency by enhancing immune function,as it could invigorate spleen and replenish Qi.The optimal dose and best ratio of Qianshi and Shanyao were expected to be 2∶1(30 g∶15 g).
