目的:探讨 ghrelin 对人卵巢癌细胞株 HO-8910增殖及凋亡作用的影响。方法将不同质量浓度的 ghre-lin(400、500、600、700、800 ng·mL-1)与人卵巢癌细胞株 HO-8910共同培养,采用 MTT 法检测细胞增殖的抑制率,Tunel 法检测细胞凋亡,蛋白免疫印迹法(Western blotting)检测磷酸化的 ERK1/2(p-ERK1/2)的表达。结果ghrelin≥600 ng·mL-1时对 HO-8910细胞增殖的抑制率具有显著作用(P <0.05)。600 ng·mL-1质量浓度的 gh-relin 作用于 HO-8910细胞20 min 后,细胞凋亡、p-ERK1/2的灰度值均显著减少(均 P <0.05)。结论ghrelin 能抑制人卵巢癌细胞株 HO-8910增殖、促进其凋亡;ERK1/2可能参与了 ghrelin 对人卵巢癌细胞株 HO-8910作用的过程。

목적:탐토 ghrelin 대인란소암세포주 HO-8910증식급조망작용적영향。방법장불동질량농도적 ghre-lin(400、500、600、700、800 ng·mL-1)여인란소암세포주 HO-8910공동배양,채용 MTT 법검측세포증식적억제솔,Tunel 법검측세포조망,단백면역인적법(Western blotting)검측린산화적 ERK1/2(p-ERK1/2)적표체。결과ghrelin≥600 ng·mL-1시대 HO-8910세포증식적억제솔구유현저작용(P <0.05)。600 ng·mL-1질량농도적 gh-relin 작용우 HO-8910세포20 min 후,세포조망、p-ERK1/2적회도치균현저감소(균 P <0.05)。결론ghrelin 능억제인란소암세포주 HO-8910증식、촉진기조망;ERK1/2가능삼여료 ghrelin 대인란소암세포주 HO-8910작용적과정。
Objective To explore the effects of ghrelin on the proliferation and apoptosis of hu-man ovarian cancer HO-8910 cells.Methods HO-8910 cells were treated with different concen-trations of ghrelin(400,500,600,700 and 800 ng·mL-1 ).Cell proliferation,apoptosis and phos-phorylated ERK1/2(p-ERK1/2)expression were measured by MTT,TUNEL and Western blot-ting assays,respectively.Results The treatment with ghrelin(600,700 and 800 ng·mL-1 )sig-nificantly inhibited the proliferation HO-8910 cells(P <0.05).Furthermore,the treatment with ghrelin(600 ng·mL-1 )for 20 minutes significantly decreased apoptosis and p-ERK1/2 grayscale value(P <0.05).Conclusion Ghrelin treatment can inhibit proliferation and promote apoptosis in human ovarian cancer HO-8910 cells.In addition,ERK1/2 may be involved in the mechanism of action of ghrelin.
