目的探讨血管紧张素转换酶抑制剂( angiotensin-converting enzyme inhibitors , ACEI )对去卵巢骨质疏松小鼠腰椎骨组织的影响。方法将96只8周龄SPF级C57 BL/6小鼠按数字表法随机分为假手术组(Sham group),模型组(OVX group)和ACEI治疗组(Treatment group),每组各32只, micro-CT检测并经三维重建获得骨组织微观结构, RT-PCR检测腰椎骨组织骨保护素( osteoprotegerin , OPG),核因子κB受体活化因子配体( receptor activator of NF-κB ligand, RANKL) mRNA的表达。结果治疗组及假手术组骨体积分数( bone volume fraction , BV/TV)、骨小梁数目( trabecular number , Tb.N)及骨小梁厚度( trabecular thickness , Tb.Th)均显著高于模型组(P<0.05),结构模型指数(structure model index, SMI)、骨小梁间隙(trabecu-lar space, Tb.Sp)及骨表面积体积比( ratio of bone surface and bone volume of trabecula , BS/BV)均显著低于模型组( P<0.05)。治疗后治疗组及假手术组骨组织中OPG mRNA表达均较模型组高( P<0.05), RANKL mRNA表达均较模型组低( P<0.05);治疗组骨组织中OPG 及RANKL mRNA表达与假手术组比较,差异无统计学意义( P>0.05)。结论血管紧张素转换酶抑制剂能部分改善去卵巢骨质疏松小鼠骨组织负转换状态,而有助于防治绝经后骨质疏松症。

목적탐토혈관긴장소전환매억제제( angiotensin-converting enzyme inhibitors , ACEI )대거란소골질소송소서요추골조직적영향。방법장96지8주령SPF급C57 BL/6소서안수자표법수궤분위가수술조(Sham group),모형조(OVX group)화ACEI치료조(Treatment group),매조각32지, micro-CT검측병경삼유중건획득골조직미관결구, RT-PCR검측요추골조직골보호소( osteoprotegerin , OPG),핵인자κB수체활화인자배체( receptor activator of NF-κB ligand, RANKL) mRNA적표체。결과치료조급가수술조골체적분수( bone volume fraction , BV/TV)、골소량수목( trabecular number , Tb.N)급골소량후도( trabecular thickness , Tb.Th)균현저고우모형조(P<0.05),결구모형지수(structure model index, SMI)、골소량간극(trabecu-lar space, Tb.Sp)급골표면적체적비( ratio of bone surface and bone volume of trabecula , BS/BV)균현저저우모형조( P<0.05)。치료후치료조급가수술조골조직중OPG mRNA표체균교모형조고( P<0.05), RANKL mRNA표체균교모형조저( P<0.05);치료조골조직중OPG 급RANKL mRNA표체여가수술조비교,차이무통계학의의( P>0.05)。결론혈관긴장소전환매억제제능부분개선거란소골질소송소서골조직부전환상태,이유조우방치절경후골질소송증。
Objective To explore the effect of angiotensin converting enzyme inhibitor ( ACEI) on lumbar bone tissue in ovariectomized mice .Methods All 96 SPF C57BL/6 mice (8 weeks old) were randomly divided into sham op-eration group ( Sham group ) , model group ( OVX group ) , and ACEI treatment group ( Treatment group ) , 32 in each group.The 3D reconstruction of the microstructure of bone were analyed by micro-CT and the expression of mRNA of os-teoprotegerin ( OPG) and receptor activator of NF-κB ligand ( RANKL) of lumbar spine were detected by reverse tran-scription-polymerase chain reaction (RT-PCR) methods.Results The bone volume fraction (BV/TV), the number of trabecula ( Tb.N) and the thickness of trabecula ( Tb.Th) in the treatment group and the sham operation group were significantly higher than model group (P<0.05); the structure model index (SMI), the trabecular space (Tb.Sp), and the ratio of bone surface and bone volume of trabecula ( BS/BV) were significantly lower than that of model group ( P<0.05 ) .The expression of OPG mRNA in the treatment group and the sham operation group were significantly higher than model group after treatment ( P<0.05 ) , and the RANKL mRNA expression were lower than the model group ( P<0.05 ) .The expression of OPG and RANKL mRNA were not significantly different between the treatment group and the sham operation group .Conclusion Angiotensin converting enzyme inhibitors can improve the high bone turnover state of ovariectomized osteoporosis mice , so can prevent and treat postmenopausal osteoporosis .
