目的:建立 HPLC‐MS测定大鼠血浆中丹酚酸B质量浓度的方法,预测盐酸右美托咪定对丹酚酸B在大鼠体内代谢的影响。方法将SD大鼠随机分成丹酚酸B组和丹酚酸B+盐酸右美托咪定组,采用 HPLC‐MS法对大鼠体内丹酚酸B血药质量浓度进行测定。结果丹酚酸B血浆加标样品在0.01~10μg · m L -1内线性关系良好( r=0.9978);丹酚酸B高、中、低质量浓度回收率均在84%~98%之间。与丹酚酸B组比,丹酚酸B+盐酸右美托咪定组中丹酚酸B清除速率(CL)有所降低,药时曲线下面积(AUC)有所增加,但均无显著性影响(P>0.05)。结论在大鼠体内盐酸右美托咪定对丹酚酸B代谢无明显影响。

목적:건립 HPLC‐MS측정대서혈장중단분산B질량농도적방법,예측염산우미탁미정대단분산B재대서체내대사적영향。방법장SD대서수궤분성단분산B조화단분산B+염산우미탁미정조,채용 HPLC‐MS법대대서체내단분산B혈약질량농도진행측정。결과단분산B혈장가표양품재0.01~10μg · m L -1내선성관계량호( r=0.9978);단분산B고、중、저질량농도회수솔균재84%~98%지간。여단분산B조비,단분산B+염산우미탁미정조중단분산B청제속솔(CL)유소강저,약시곡선하면적(AUC)유소증가,단균무현저성영향(P>0.05)。결론재대서체내염산우미탁미정대단분산B대사무명현영향。
Objective To establish an HPLC‐MS method for the determination of salvianolic acid B in rat plasma and to obtain the in‐formation about the influence of dexmedetomidine hydrochloride on the metabolism of salvianolic acid B in vivo .Methods The rats were divided into salvianolic acid B group and salvianolic acid B+dexmedetomidine hydrochloride group randomly .Plasma concen‐tration of salvianolic acid B was determinated by HPLC‐MS after oral administration .Results The calibration curve was linear in the range of 0 .01‐10 .00 μg · mL -1 for salvianolic acid B (r=0 .997 8) .The average recoveries of salvianolic acid B at high ,medi‐um ,and low concentration were between 84% and 98% .Compared with salvianolic acid B group ,in salvianolic acid B+ dexme‐detomidine hydrochloride group ,clearance rate (CL) decreased and area under the curve (AUC) increased slightly ,but there was no significant difference(P> 0 .05) .Conclusion Dexmedetomidine hydrochloride had no effect on the metabolism of salvianolic acid B in rat .No drug‐interaction was observed.
