目的:探讨负载MB49细胞粗提全抗原的树突状细胞(DC2.4)疫苗对C57BL/6小鼠膀胱癌MB49皮下移植瘤模型的抑瘤效果。方法制备负载MB49细胞粗提全抗原的DC疫苗;建立小鼠MB49膀胱癌皮下移植瘤模型。造模后第7 d、14 d,疫苗组小鼠于右前腋皮下注射疫苗,PBS对照组相应注射PBS,正常组正常饲养。造模第21 d,测量肿瘤体积和重量。应用免疫组化染色方法检测肿瘤组织内CD4+、CD8+细胞数。应用SPSS13.0统计分析测量数据。结果 DC疫苗组和PBS对照组小鼠肿瘤体积随时间呈现不同程度增长趋势,DC疫苗组缓于PBS对照组,差异有统计学意义(P<0.01);造模21 d,DC疫苗组小鼠肿瘤重量小于PBS对照组,差异有统计学意义(P<0.01);免疫组化染色检测小鼠肿瘤组织内CD4+、CD8+T细胞数,DC疫苗组CD4+、CD8+T细胞数高于PBS对照组,差异有统计学意义(P<0.01)。结论 DC2.4树突状细胞疫苗抑制了模型小鼠肿瘤的生长,起到了抗肿瘤的免疫学效应。

Objective To investigate the antitumor effect of dendritic cell vaccine against C57BL/6 cells loaded with MB49 cells in MB49 mice. Methods DC vaccine against MB49 cells were prepared by loading MB49 cells, and the model was established in mice. After making the model, the 7 d, 14 d, the vaccine were injected in right anterior axillary of vaccine group mice, and the mice of PBS control group were injected with PBS, normal feeding to normal group. 21 d after modeling, measurement of the volume and weight of the tumor. The number of CD4+ and CD8+ cells in tumor tissues was detected by immunohistochemical staining. SPSS13.0 statistical analysis of measurement data. Results The tumor volume of DC vaccine group and PBS control group showed different degrees of growth trend, DC vaccine group was signiifcantly slower than that in 21 d control group (P<0.01). The tumor weight was signiifcantly less than that in DC group (P<0.01). In tumor tissue of CD4+and CD8+T cell number and DC vaccine group of CD4+, CD8+T cel number higher than the PBS control group. The difference was statistical y signiifcant (P<0.01). Conclusion DC2.4 dendritic cel vaccine inhibits tumor growth in mouse model, which has the effect of anti-tumor.
